In the 40 mg Cohort, Two Patients Achieved 15 Hours of Controlled Fasting, Improvements in Glucose Control and No Longer Required Starch Supplementation Following Treatment

Safety Profile of BEAM-301 Consistent with LNP-Based Therapies

Beam Plans to Engage FDA on Potential Path to Approval; Enrollment in 60 mg Dose Cohort Underway

Company to Host Investor Webcast Today, October 27, 2026, at 8:00 a.m. ET

CAMBRIDGE, Mass., Oct. 27, 2026 (GLOBE NEWSWIRE) — Beam Therapeutics Inc. (Nasdaq: BEAM), a biotechnology company developing precision genetic medicines through base editing, today reported initial data from the ongoing Phase 1/2 clinical trial evaluating BEAM-301 for the treatment of glycogen storage disease type Ia (GSDIa). The data will be presented during a poster session at the 33rd Annual Congress of the European Society of Gene and Cell Therapy (ESGCT) in Hamburg, Germany. BEAM-301 is an investigational liver-targeting lipid-nanoparticle (LNP) formulation of base editing reagents designed to correct the R83C mutation, one of the most prevalent disease-causing mutations in GSDIa and one that is associated with the most severe form of the disease.

“These findings demonstrate the potential for BEAM-301 to fundamentally change how patients with GSDIa manage their disease,” said Rebecca Riba-Wolman, M.D., director of the glycogen storage disease program and disorders of hypoglycemia at Connecticut Children’s and University of Connecticut School of Medicine, and presenting investigator in the BEAM-301 Phase 1/2 trial. “The ability of two patients to achieve 15 hours of controlled fasting and no longer require starch supplementation following a single treatment represents a significant departure from the intensive daily management typically required for this disease. While these are early results, the improvements in glucose regulation and reductions in starch requirements provide encouraging evidence of BEAM-301’s potential to deliver meaningful and lasting benefits for patients.”

BEAM-301 Phase 1/2 Clinical Trial Data Update
BEAM-301 is being evaluated in an open-label Phase 1/2 dose-exploration trial to investigate its safety, tolerability, pharmacodynamics, pharmacokinetics and efficacy in patients with GSDIa who are homozygous or compound heterozygous for the R83C variant. As of the August 17, 2026, data cutoff, six patients had received a single intravenous infusion of BEAM-301, including three patients treated with 20 mg and three patients treated with 40 mg. Median follow-up across both cohorts was 7.6 months (range: 1.9–16.6 months).

Key highlights include:

  • Safety Profile Consistent with LNP-Based Therapies: BEAM-301 demonstrated an acceptable safety profile consistent with LNP-based therapies, with no dose-limiting toxicities, Grade 4 or 5 treatment-emergent adverse events (TEAEs), or treatment discontinuations due to TEAEs reported. All patients experienced at least one TEAE considered related to BEAM-301. Four of six patients experienced serious TEAEs considered related to BEAM-301, including three Grade 3 infusion-related reactions (IRRs). Premedication with shorter-acting steroids, which are typically used in this patient population, may have contributed to the Grade 3 IRRs observed. The steroid premedication and infusion rate regimens have since been modified. Two other serious TEAEs of diarrhea and rhabdomyolysis occurred in two patients; both have resolved. With the exception of a single Grade 2 elevation in aspartate aminotransferase (AST), all transaminase elevations were Grade 1. All liver enzyme elevations resolved without treatment, and total bilirubin remained within normal limits.
  • Restoration of Endogenous Glucose Production Following a Single Treatment: BEAM-301 demonstrated dose-dependent improvements in time to fasting hypoglycemia (TTH), a key measure of glucose regulation in patients with GSDIa. Median TTH increased from 2.9 hours at baseline to 4.3 hours at Day 36 in the 20 mg cohort, and from 2.5 hours at baseline to 15.0 hours at Day 36 in the 40 mg cohort. Two of three patients in the 40 mg cohort achieved the protocol-defined maximum controlled fasting duration of 15 hours, consistent with restored endogenous glucose production. During the 15-hour fasting challenges, blood glucose levels were generally maintained within the euglycemic range. One patient in the 40 mg cohort did not show an increase in TTH at the first post-dose assessment; several factors may have contributed, including elevated body mass index (BMI) and heterozygosity for the R83C mutation.
  • 100% Reduction in Daily Starch Supplementation Observed in Two Patients: Daily starch intake and daily number of starch doses generally decreased following treatment with BEAM-301 (baseline: 6–7 and 5–6 doses/day in the 20 mg and 40 mg cohorts, respectively), with two of three patients in the 40 mg cohort achieving a 100% reduction in daily starch requirements and no longer requiring starch supplementation at their latest reported assessments.
  • Improvements in Glucose Control Despite Reduced Starch Intake: Continuous glucose monitoring demonstrated increased time spent in the euglycemic range and decreased time in the hyperglycemic range following treatment with BEAM-301, despite reductions in daily starch intake. Patients generally did not experience an increase in time spent in hypoglycemia. Triglyceride levels were generally comparable to baseline at the latest evaluable time point.

Based on the safety and clinical findings observed to date, Beam is advancing BEAM-301 into the 60 mg dose-escalation cohort, with enrollment currently underway. Beam also plans to engage with the U.S. Food and Drug Administration (FDA) regarding a potential path to approval.

“The promise of base editing is to deliver transformative outcomes that correct the root cause of disease with a one-time therapy. These initial results show that we are continuing to advance toward that vision, generating important clinical proof of concept for BEAM-301 in GSDIa and once again demonstrating the potential of base editing to fundamentally change the treatment paradigm for patients,” said John Evans, chief executive officer of Beam Therapeutics. “These findings also reinforce the strength of our fully integrated platform, combining our proprietary base editing technology, validated delivery capabilities, and internal manufacturing to enable the predictable development and rapid production of precision genetic medicines. We look forward to building on these encouraging results as we advance BEAM-301 into the next dose cohort, engage with regulators on a potential pathway to approval, and progress our broader pipeline.”

ESGCT Investor Event Information
Beam will host a live and webcast investor event today, Tuesday, October 27, at 8:00 a.m. ET to review the data presented at this year’s ESGCT congress. The event will be webcast live and can be accessed under “Events & Presentations” in the Investors section of the company’s website at www.beamtx.com. The archived webcast will be available on the company’s website beginning approximately two hours after the event.

About BEAM-301
BEAM-301 is an investigational liver-targeting lipid-nanoparticle (LNP) formulation of base editing reagents designed to correct the R83C mutation, one of the most prevalent disease-causing mutations and one that results in the most severe form of glycogen storage disease type Ia (GSDIa). BEAM-301 is being evaluated in a Phase 1/2, open-label, dose-exploration and dose-expansion clinical trial to investigate its safety, tolerability, pharmacodynamics, pharmacokinetics and efficacy in patients with GSDIa.

About Glycogen Storage Disease Type Ia (GSDIa)
GSDIa is an autosomal recessive disorder caused by mutations in the G6PC1 gene that disrupt a key enzyme, glucose-6-phosphatase (G6Pase), critical for maintaining glucose homeostasis. Inhibition of G6Pase activity results in low fasting blood glucose levels that can result in seizures and death. Patients with the G6PC1 R83C mutation typically require frequent cornstarch administration, without which they may enter into hypoglycemic shock within one to three hours. There remains significant unmet need for patients with GSDIa.

About Beam Therapeutics
Beam Therapeutics (Nasdaq: BEAM) is a biotechnology company committed to establishing the leading, fully integrated platform for precision genetic medicines. To achieve this vision, Beam has assembled a platform with integrated gene editing, delivery and internal manufacturing capabilities. Beam’s suite of gene editing technologies is anchored by base editing, a proprietary technology that is designed to enable precise, predictable and efficient single base changes, at targeted genomic sequences, without making double-stranded breaks in the DNA. This has the potential to enable a wide range of therapeutic editing strategies that Beam is using to advance a diversified portfolio of base editing programs. Beam is a values-driven organization committed to its people, cutting-edge science, and a vision of providing lifelong cures to patients suffering from serious diseases.

CALGARY, Alberta, Oct. 27, 2026 (GLOBE NEWSWIRE) — Questor Technology Inc. (“Questor” or the “Company”) (TSX-V: QST) announces the appointment of Audrey Mascarenhas, F.C.A.E., P.Eng., as President and Chief Executive Officer, effective immediately. Ms. Mascarenhas succeeds Mike Lindsay, who served as Interim President and Chief Executive Officer since April 2026.

Ms. Mascarenhas has been with Questor since 1999 and has spent more than 25 years developing and commercializing its clean combustion technology and continues to grow its role in effective emissions reduction for the oil and gas, energy, industrial, and municipal sectors. She served as President and Chief Executive Officer from 2005 to April 2026. Before Questor, she spent 17 years at Gulf Canada Resources (1982–1999) in upstream exploration and corporate transactions, giving her more than four decades of experience in the energy industry. She is a professional engineer with deep experience in commercializing clean technology, serving international energy markets, and TSXV governance and disclosure, including her board experience at TMX Group Limited.

“Questor’s technology has a strong track record in achieving unprecedented performance, and interest from our global and local clients remains high. My priority is to bring value-add solutions to our clients, with disciplined execution and a clear focus on clients who value what we do. I am grateful to the shareholders and employees who continue to support the Company, and I look forward to getting back to work,” said Ms. Mascarenhas.

The Company’s annual general meeting, originally scheduled for June 24, 2026, was rescheduled to June 15, 2026, and subsequently postponed despite indications from the proxy tally that shareholders wanted a change. The meeting was finally held on September 9, 2026, and shareholders again voted for a change in Board composition. The Company incurred significant costs associated with the proxy contest and related proceedings. The Company’s new Board is aligned with and invested in Questor’s success and, with the leadership transition complete, is focused on disciplined execution, commercial growth and long-term value for shareholders.

Questor now moves forward with renewed focus on its global and local opportunities. Discussions with Tier 1 clients have resumed following Ms. Mascarenhas’ return, and the Board and management believe the Company is well positioned to convert this renewed engagement into long-term growth for shareholders.

TONMYA improved Fibromyalgia Impact Questionnaire-Revised (FIQR) symptoms and function scores versus placebo in both domains at Week 14 in RELIEF and RESILIENT Phase 3 pivotal studies

Tonix commercially launched TONMYA® in November 2025, the first fibromyalgia drug for adults in the U.S. in over 15 years approved by the U.S. Food and Drug Administration (FDA)

BERKELEY HEIGHTS, N.J., Oct. 27, 2026 (GLOBE NEWSWIRE) — Tonix Pharmaceuticals Holding Corp. (Nasdaq: TNXP) (“Tonix” or the “Company”), a fully integrated, commercial-stage biopharmaceutical company, presented data from an analysis of the Phase 3 RELIEF and RESILIENT studies of TONMYA® (cyclobenzaprine HCl sublingual tablets) for the treatment of fibromyalgia in adults demonstrating outcomes as measured by the Fibromyalgia Impact Questionnaire-Revised (FIQR) at the International Association for the Study of Pain (IASP) 2026 World Congress on Pain, held October 26-30, 2026, in Bangkok, Thailand.

“TONMYA improved a broad spectrum of physical symptoms and functional capacity in adults with fibromyalgia in two Phase 3 studies,” said Seth Lederman, M.D., Chief Executive Officer of Tonix Pharmaceuticals. “The core physical symptoms of fibromyalgia can be debilitating for patients with fibromyalgia, interfering with their quality of life. We believe the data reinforce the role of TONMYA as a first-line, non-opioid treatment option that can help patients function better in their daily lives.”

RELIEF and RESILIENT were randomized, double-blind, placebo-controlled Phase 3 trials that assessed the efficacy and safety of TONMYA versus placebo over 14 weeks in adults with fibromyalgia who were diagnosed with the condition diagnosed according to the 2016 American College of Rheumatology (ACR) diagnostic criteria. In both trials, participants were randomized 1:1 to receive either TONMYA (2.8 mg tablets at bedtime for two weeks, followed by 5.6 mg at bedtime through Week 14) or matching placebo for 14 weeks.

RELIEF and RESILIENT evaluated the same primary efficacy and key secondary efficacy endpoints. The primary efficacy endpoint was the change from baseline in the weekly average of daily self-reported numeric rating scale (NRS) pain scores (0-10) at Week 14. There were multiple key secondary efficacy endpoints, including change from baseline in the FIQR Symptoms domain score and change from baseline in the FIQR Function domain score. The FIQR assesses overall fibromyalgia burden and severity over the prior seven days, including physical functioning and symptoms. FIQR assessments were conducted at Weeks 2, 6, 10, and 14, with Week 14 as the pre-specified timepoint of interest.

In the RELIEF and RESILIENT studies, TONMYA provided statistically and clinically significant improvements over placebo in the change from baseline in the weekly average of daily numeric rating scale pain scores versus placebo at Week 14. A greater reduction from baseline in both the FIQR Symptoms and FIQR Function total scores was seen in participants who received TONMYA as compared to those who received placebo at Weeks 2, 6, 10, and 14 in both studies. Week 14 was statistically significant (p≤0.001) for both domains in RESILIENT and nominally significant (due to the hierarchical testing of endpoints used to control type 1 error) in RELIEF (p<0.01). TONMYA was generally well tolerated, with an adverse event (AE) profile consistent with previous studies; no new safety signals were observed. The most common treatment-emergent adverse events (TEAEs) were local oral administration site reactions.

A copy of the Company’s poster presentation, “Cyclobenzaprine HCl Sublingual Reduces Fibromyalgia Impact on Physical Symptoms & Functional Ability,” is available under the Scientific Presentations tab on the Tonix website at https://www.tonixpharma.com/scientific-presentations/.

Perform a pregnancy test prior to initiation of treatment with TONMYA to exclude use of TONMYA during the first trimester of pregnancy.

Serotonin syndrome: Concomitant use of TONMYA with selective serotonin reuptake inhibitors (SSRIs), serotonin norepinephrine reuptake inhibitors (SNRIs), tricyclic antidepressants, tramadol, bupropion, meperidine, verapamil, or MAO inhibitors increases the risk of serotonin syndrome, a potentially life-threatening condition. Serotonin syndrome symptoms may include mental status changes, autonomic instability, neuromuscular abnormalities, and/or gastrointestinal symptoms. Treatment with TONMYA and any concomitant serotonergic agent should be discontinued immediately if serotonin syndrome symptoms occur and supportive symptomatic treatment should be initiated. If concomitant treatment with TONMYA and other serotonergic drugs is clinically warranted, careful observation is advised, particularly during treatment initiation or dosage increases.

Tricyclic antidepressant-like adverse reactions: Cyclobenzaprine is structurally related to TCAs. TCAs have been reported to produce arrhythmias, sinus tachycardia, prolongation of the conduction time leading to myocardial infarction and stroke. If clinically significant central nervous system (CNS) symptoms develop, consider discontinuation of TONMYA. Caution should be used when TCAs are given to patients with a history of seizure disorder, because TCAs may lower the seizure threshold. Patients with a history of seizures should be monitored during TCA use to identify recurrence of seizures or an increase in the frequency of seizures.

Atropine-like effects: Use with caution in patients with a history of urinary retention, angle-closure glaucoma, increased intraocular pressure, and in patients taking anticholinergic drugs.

CNS depression and risk of operating a motor vehicle or hazardous machinery: TONMYA monotherapy may cause CNS depression. Concomitant use of TONMYA with alcohol, barbiturates, or other CNS depressants may increase the risk of CNS depression. Advise patients not to operate a motor vehicle or dangerous machinery until they are reasonably certain that TONMYA therapy will not adversely affect their ability to engage in such activities. Oral mucosal adverse reactions: In clinical studies with TONMYA, oral mucosal adverse reactions occurred more frequently in patients treated with TONMYA compared to placebo. Advise patients to moisten the mouth with sips of water before administration of TONMYA to reduce the risk of oral sensory changes (hypoesthesia). Consider discontinuation of TONMYA if severe reactions occur.

ADVERSE REACTIONS

The most common adverse reactions (incidence ≥2% and at a higher incidence in TONMYA-treated patients compared to placebo-treated patients) were oral hypoesthesia, oral discomfort, abnormal product taste, somnolence, oral paresthesia, oral pain, fatigue, dry mouth, and aphthous ulcer.

DRUG INTERACTIONS

MAO inhibitors: Life-threatening interactions may occur.

Other serotonergic drugs: Serotonin syndrome has been reported.

CNS depressants: CNS depressant effects of alcohol, barbiturates, and other CNS depressants may be enhanced.

Tramadol: Seizure risk may be enhanced.

Guanethidine or other similar acting drugs: The antihypertensive action of these drugs may be blocked.

USE IN SPECIFIC POPULATIONS

Pregnancy: Based on animal data, TONMYA may cause fetal harm when administered to a pregnant woman. The limited amount of available observational data on oral cyclobenzaprine use in pregnancy is of insufficient quality to inform a TONMYA-associated risk of major birth defects, miscarriage, or adverse maternal or fetal outcomes. Advise pregnant women about the potential risk to the fetus with maternal exposure to TONMYA and to avoid use of TONMYA two weeks prior to conception and through the first trimester of pregnancy. Report pregnancies to the Tonix Medicines, Inc., adverse-event reporting line at 1-888-869-7633 (1-888-TNXPMED).

Lactation: A small number of published cases report the transfer of cyclobenzaprine into human milk in low amounts, but these data cannot be confirmed. There are no data on the effects of cyclobenzaprine on a breastfed infant, or the effects on milk production. The developmental and health benefits of breastfeeding should be considered along with the mother’s clinical need for TONMYA and any potential adverse effects on the breastfed child from TONMYA or from the underlying maternal condition.

Pediatric use: The safety and effectiveness of TONMYA have not been established.

Geriatric patients: Of the total number of TONMYA-treated patients in the clinical trials in adult patients with fibromyalgia, none were 65 years of age and older. Clinical trials of TONMYA did not include sufficient numbers of patients 65 years of age and older to determine whether they respond differently from younger adult patients.

Hepatic impairment: The recommended dosage of TONMYA in patients with mild hepatic impairment (HI) (Child Pugh A) is 2.8 mg once daily at bedtime, lower than the recommended dosage in patients with normal hepatic function. The use of TONMYA is not recommended in patients with moderate HI (Child Pugh B) or severe HI (Child Pugh C). Cyclobenzaprine exposure (AUC) was increased in patients with mild HI and moderate HI compared to subjects with normal hepatic function, which may increase the risk of TONMYA-associated adverse reactions.
