(Nasdaq: TNXP) (“Tonix” or “the Company”), a fully integrated, commercial-stage biopharmaceutical company, today announced a second major managed Medicare agreement for TONMYA® (cyclobenzaprine HCl sublingual tablets) for the treatment of fibromyalgia in adults that is effective immediately, adding approximately 21 million Medicare lives (38% of approximately 55 million Medicare lives in the U.S.). Medicare coverage now totals 32 million Medicare lives in the U.S. (58% of approximately 55 million Medicare lives in the U.S.).

Total commercial and government payer coverage for TONMYA now reaches approximately 166 million covered lives (53% of approximately 314 million total U.S. lives). Previously, Tonix announced commercial payer agreements with two leading group purchasing organizations, and one major managed Medicare agreement. In addition, TONMYA is covered by Medicaid in nearly all states.

INDICATION

TONMYA is indicated for the treatment of fibromyalgia in adults.

CONTRAINDICATIONS

TONMYA is contraindicated:

In patients with hypersensitivity to cyclobenzaprine or any inactive ingredient in TONMYA. Hypersensitivity reactions may manifest as an anaphylactic reaction, urticaria, facial and/or tongue swelling, or pruritus. Discontinue TONMYA if a hypersensitivity reaction is suspected. With concomitant use of monoamine oxidase (MAO) inhibitors or within 14 days after discontinuation of an MAO inhibitor. Hyperpyretic crisis seizures and deaths have occurred in patients who received cyclobenzaprine (or structurally similar tricyclic antidepressants) concomitantly with MAO inhibitors drugs.

During the acute recovery phase of myocardial infarction, and in patients with arrhythmias, heart block or conduction disturbances, or congestive heart failure. In patients with hyperthyroidism.

WARNINGS AND PRECAUTIONS

Embryofetal toxicity: Based on animal data, TONMYA may cause neural tube defects when used two weeks prior to conception and during the first trimester of pregnancy. Advise females of reproductive potential of the potential risk and to use effective contraception during treatment and for two weeks after the final dose. Perform a pregnancy test prior to initiation of treatment with TONMYA to exclude use of TONMYA during the first trimester of pregnancy.

Serotonin syndrome: Concomitant use of TONMYA with selective serotonin reuptake inhibitors (SSRIs), serotonin norepinephrine reuptake inhibitors (SNRIs), tricyclic antidepressants, tramadol, bupropion, meperidine, verapamil, or MAO inhibitors increases the risk of serotonin syndrome, a potentially life-threatening condition. Serotonin syndrome symptoms may include mental status changes, autonomic instability, neuromuscular abnormalities, and/or gastrointestinal symptoms. Treatment with TONMYA and any concomitant serotonergic agent should be discontinued immediately if serotonin syndrome symptoms occur and supportive symptomatic treatment should be initiated. If concomitant treatment with TONMYA and other serotonergic drugs is clinically warranted, careful observation is advised, particularly during treatment initiation or dosage increases.

Tricyclic antidepressant-like adverse reactions: Cyclobenzaprine is structurally related to TCAs. TCAs have been reported to produce arrhythmias, sinus tachycardia, prolongation of the conduction time leading to myocardial infarction and stroke. If clinically significant central nervous system (CNS) symptoms develop, consider discontinuation of TONMYA. Caution should be used when TCAs are given to patients with a history of seizure disorder, because TCAs may lower the seizure threshold. Patients with a history of seizures should be monitored during TCA use to identify recurrence of seizures or an increase in the frequency of seizures.

Atropine-like effects: Use with caution in patients with a history of urinary retention, angle-closure glaucoma, increased intraocular pressure, and in patients taking anticholinergic drugs.

CNS depression and risk of operating a motor vehicle or hazardous machinery: TONMYA monotherapy may cause CNS depression. Concomitant use of TONMYA with alcohol, barbiturates, or other CNS depressants may increase the risk of CNS depression. Advise patients not to operate a motor vehicle or dangerous machinery until they are reasonably certain that TONMYA therapy will not adversely affect their ability to engage in such activities. Oral mucosal adverse reactions: In clinical studies with TONMYA, oral mucosal adverse reactions occurred more frequently in patients treated with TONMYA compared to placebo. Advise patients to moisten the mouth with sips of water before administration of TONMYA to reduce the risk of oral sensory changes (hypoesthesia). Consider discontinuation of TONMYA if severe reactions occur.

ADVERSE REACTIONS

The most common adverse reactions (incidence ≥2% and at a higher incidence in TONMYA-treated patients compared to placebo-treated patients) were oral hypoesthesia, oral discomfort, abnormal product taste, somnolence, oral paresthesia, oral pain, fatigue, dry mouth, and aphthous ulcer.

DRUG INTERACTIONS

MAO inhibitors: Life-threatening interactions may occur.

Other serotonergic drugs: Serotonin syndrome has been reported.

CNS depressants: CNS depressant effects of alcohol, barbiturates, and other CNS depressants may be enhanced.

Tramadol: Seizure risk may be enhanced.

Guanethidine or other similar acting drugs: The antihypertensive action of these drugs may be blocked.

USE IN SPECIFIC POPULATIONS

Pregnancy: Based on animal data, TONMYA may cause fetal harm when administered to a pregnant woman. The limited amount of available observational data on oral cyclobenzaprine use in pregnancy is of insufficient quality to inform a TONMYA-associated risk of major birth defects, miscarriage, or adverse maternal or fetal outcomes. Advise pregnant women about the potential risk to the fetus with maternal exposure to TONMYA and to avoid use of TONMYA two weeks prior to conception and through the first trimester of pregnancy. Report pregnancies to the Tonix Medicines, Inc., adverse-event reporting line at 1-888-869-7633 (1-888-TNXPMED).

Lactation: A small number of published cases report the transfer of cyclobenzaprine into human milk in low amounts, but these data cannot be confirmed. There are no data on the effects of cyclobenzaprine on a breastfed infant, or the effects on milk production. The developmental and health benefits of breastfeeding should be considered along with the mother’s clinical need for TONMYA and any potential adverse effects on the breastfed child from TONMYA or from the underlying maternal condition.

Pediatric use: The safety and effectiveness of TONMYA have not been established.

Geriatric patients: Of the total number of TONMYA-treated patients in the clinical trials in adult patients with fibromyalgia, none were 65 years of age and older. Clinical trials of TONMYA did not include sufficient numbers of patients 65 years of age and older to determine whether they respond differently from younger adult patients.

Hepatic impairment: The recommended dosage of TONMYA in patients with mild hepatic impairment (HI) (Child Pugh A) is 2.8 mg once daily at bedtime, lower than the recommended dosage in patients with normal hepatic function. The use of TONMYA is not recommended in patients with moderate HI (Child Pugh B) or severe HI (Child Pugh C). Cyclobenzaprine exposure (AUC) was increased in patients with mild HI and moderate HI compared to subjects with normal hepatic function, which may increase the risk of TONMYA-associated adverse reactions.

Evenamide – advancing schizophrenia treatment

Newron continued to make substantial progress with its pivotal ENIGMA-TRS Phase 3 development program during the first half of 2026. The program evaluates evenamide as an add-on therapy in patients with treatment-resistant schizophrenia (TRS), a population that remains underserved by currently available treatments.

ENIGMA-TRS 1 is an international, 52-week, randomized, double-blind, placebo-controlled Phase 3 study evaluating the efficacy, tolerability and safety of the 15mg BID and 30mg BID doses of evenamide compared with placebo. The study is designed to enroll at least 600 patients at centers across Europe, Asia, Latin America and Canada.

Post-period, in September 2026, the last of 996 patients entered screening for enrollment. Newron expects target enrollment of at least 600 patients to be reached around mid-October 2026.

The primary efficacy endpoint is the change from baseline in the Positive and Negative Syndrome Scale (PANSS) score at 12 weeks. Newron expects to announce Topline 12-week results in Q1 2027. The study will continue under double-blind, placebo-controlled conditions through 52 weeks.

ENIGMA-TRS 2 is a 12-week, randomized, double-blind, placebo-controlled Phase 3 study designed to enroll at least 400 patients and evaluate the efficacy, tolerability and safety of the 15mg BID dose of evenamide.

Enrollment started in the US in December 2025, followed by regulatory approvals in study centers in Asia and Latin America. In April 2026, Newron announced a pause in enrollment at US study centers following notification to the FDA of the sudden death of a study participant at a clinical site outside the US, in accordance with standard practice.

Following a constructive Type A meeting with the FDA in July, Newron is implementing changes discussed with the FDA and expects U.S. enrollment to resume following FDA clearance. Study centers outside the US are not affected by the pause.

In January 2026, Newron’s partner EA Pharma (part of the Eisai Group), announced initiation of its Phase 3 clinical trial with evenamide in Japan.

Intellectual property and scientific engagement

In January 2026, the European Patent Office issued a decision to grant Newron an additional Composition of Matter patent providing protection for evenamide through 2044. The patent covers crystalline forms of evenamide, processes for their preparation and their uses, further strengthening the Company’s intellectual property position.

In February, peer-reviewed data was published in Therapeutic Advances in Psychopharmacology. The publication brought together findings from multiple randomized clinical trials and provided further scientific and mechanistic support for evenamide as a novel treatment approach for patients with schizophrenia who have an inadequate response to existing treatments, including those with TRS.

Newron also continued to engage with the scientific and investment communities at key industry and investor conferences during the reporting period.

Xadago®/safinamide – Parkinson’s disease

In partnership with Zambon and Supernus, Newron continues to further develop and market Xadago®/safinamide.

In the US, generic versions of safinamide mesylate are not permitted to launch earlier than December 2027. In the European Union, Newron expects Supplementary Protection Certificates in key markets following completion of ongoing procedures.

Corporate developments

In February, Newron entered into an agreement with existing and new shareholders from Europe and Asia for the subscription of newly issued shares for proceeds of up to EUR 38 million. To date, EUR 20.5 million has been subscribed under the agreement, including a EUR 5.5 million tranche completed post-period in September. The financing further strengthens the Company’s financial position and supports continued execution of the ENIGMA-TRS Phase 3 program.

A further EUR 5.5 million tranche from these investors is expected no later than November 30, 2026, with an additional EUR 12 million expected following disclosure of the ENIGMA-TRS results, conditional upon such results being positive.

In March, Newron announced an agreement with the European Investment Bank extending the maturity date of all outstanding tranches under its 2018 Finance Contract to June 28, 2028.

Post-period, Newron also received a further EUR 5.5 million milestone payment from EA Pharma (Eisai Group), reflecting continued progress in the development of evenamide.

At the Annual General Meeting in April, George Garibaldi and Paolo Zocchi were elected as new independent non-executive Board members, succeeding Patrick Langlois and Luca Benatti.

Financial key takeaways H1 2026

  • For the first six months of 2026, Newron reported revenues of EUR 3.2 million, compared to EUR 11.9 million in the same period of 2025. H1 2025 revenues reflect the achievement of the first milestone under the license agreement with EA Pharma (Eisai Group) and the upfront payment received from Myung In Pharm.
  • Newron’s R&D expenses increased to EUR 14.6 million in H1 2026, up from EUR 6.1 million in H1 2025.
  • This resulted in a net loss of EUR 16.4 million, compared to EUR 0.1 million in the same period in 2025.
  • Operating activities absorbed EUR 10.8 million while as of June 2025 they generated EUR 33.4 million of cash.
  • Cash and Other current financial assets as at June 30, 2026, were at EUR 32.9 million, compared to EUR 28.9 million as at December 31, 2025. These results confirm strong high-grade continuity along the Boumadine Main Trend and support the potential for continued resource growth.

Boumadine Main Trend (5.4km)

  • Multiple additional high-grade intercepts:
    • BOU-DD26-848 intercepted 392 grams per tonne (“g/t”) silver equivalent (“AgEq”) over 24.3 metres (“m”) (0.61 g/t gold (“Au”), 239 g/t silver (“Ag”), 2.6% zinc (“Zn”), and 1.9% lead (“Pb”), including 776 g/t AgEq over 10.0m (1.08 g/t Au, 488 g/t Ag, 5.2% Zn, and 3.9% Pb).
    • BOU-DD26-883 intercepted 593 g/t AgEq over 15.2m (1.05 g/t Au, 319 g/t Ag, 5.3% Zn and 3.5% Pb), including 1,038 g/t AgEq over 4.2m (1.81 g/t Au, 586g/t Ag, 8.1% Zn and 6.2% Pb).
    • BOU-DD26-0950 intercepted 299 g/t AgEq over 15.1m (1.70 g/t Au, 98 g/t Ag, 1.7% Zn and 0.6% Pb).

2026 Exploration Results

This year, 406 diamond drill holes (“DDH”), totaling 145,719m have been completed at Boumadine (Figure 1 and Appendix 2). Drilling was done on strike along the Main Trend, Tizi and Imariren. All results have been received for drill holes up to BOU-DD26-1028 (Table 1, Figure 2, Figure 3 and Appendix 1).

Today’s results, including BOU-DD26-848 and BOU-DD26-883, confirm the high-grade nature and continuity of the Boumadine Main Trend, which remains open in all directions.

Mineralization within the Boumadine Main Trend measures up to 4m wide (locally reaching over a 10m width) and is N340-oriented massive sulphide lenses/veins sharply dipping eastward (> 70°). The massive sulphide veins (>80%) are mainly composed of pyrite, with variable proportions of sphalerite, galena, and chalcopyrite. Tizi and Imariren share the same characteristics except for their N000 orientation.

Next Steps

Infill drilling, following the latest positive preliminary economic assessment (“PEA”), will be ongoing for the next 15 months with a drill program of approximately 360,000m planned.

Significant potential exists to expand the Boumadine Main Trend (currently 5.4 km), the Tizi Zone (2.0 km) and the Imariren Zone (1.2 km), with all three trends open in all directions. Follow-up drilling is also planned on the 8 km Asirem trend for later this year.

Most drilling will continue to focus on the Main Trend, Imariren and Tizi to extend known mineralization along strike and at depth, while infilling key areas to advance the Project toward a Feasibility Study (“FS”). The balance of the 2026 program (~20,000m) will target Asirem follow-up and greenfield exploration, testing geological hypotheses and targets generated over the past four years. Ongoing geological work will guide additional development priorities.

Technical Information

Aya has implemented a quality control program to comply with best practices in sampling and analysis of drill core and RC chips. For core drilling, all individual samples represent approximately one metre in length of core, which is halved. Half of the core is kept on site for reference, and its counterpart is sent for preparation and assaying to African Laboratory for Mining and Environment (“Afrilab”) in Marrakech, Morocco. For drilling using RC, all individual samples represent 1.0m in length and a representative portion is kept for every metre in some chip trays stored on site. A split samples representing 1/16th, ranging from 2 to 4 kilogram is sent for preparation and assaying to African Laboratory for Mining and Environment (“Afrilab”) in Marrakech, Morocco.

All samples are analyzed for silver, copper, iron, lead, zinc, tin, and molybdenum using Aqua regia and finished by atomic absorption spectroscopy (“AAS”). Samples grading above 200 g/t Ag are reanalyzed using fire assaying. Gold is assayed by fire assaying. Standards of different grades and blanks were inserted every 20 samples in addition to the standards, blanks and pulp duplicate inserted by Afrilab.

HAMPTON, N.J., Sept. 22, 2026 (GLOBE NEWSWIRE) — Celldex (NASDAQ:CLDX) today reported positive topline results from the Company’s Phase 3 EMBARQ-CSU1 and EMBARQ-CSU2 trials evaluating two doses of barzolvolimab in patients with chronic spontaneous urticaria (CSU) whose symptoms are inadequately controlled by H1-antihistamines. The EMBARQ-CSU1 and EMBARQ-CSU2 studies are the largest program conducted in antihistamine refractory CSU, including patients with advanced therapy experienced/refractory CSU. Both Phase 3 trials met the primary endpoint of mean change from baseline in weekly urticaria activity score (UAS7) at Week 12, and all key secondary endpoints, demonstrating clinically meaningful and statistically significant improvements in disease activity. Barzolvolimab was well-tolerated through the 24 week placebo controlled treatment period with a favorable safety profile consistent with prior Phase 2 experience. The EMBARQ-CSU trials are ongoing, with treatment continuing through 52 weeks. The Company intends to present the EMBARQ-CSU data at an upcoming medical meeting and plans to submit a Biologics License Application (BLA) to the FDA in 2027.

The Phase 3 Program is designed to establish the efficacy and safety of barzolvolimab in adult patients with CSU who remain symptomatic despite H1 antihistamine treatment. Both Phase 3 trials are randomized, double-blind, placebo-controlled, parallel group, global studies. 1,939 patients were randomized (n=963 EMBARQ-CSU1; n=976 EMBARQ-CSU2) evenly to barzolvolimab 150 mg every 4 weeks (following 300 mg loading dose), barzolvolimab 300 mg every 8 weeks (following 450 mg loading dose) for 52 weeks or placebo for 24 weeks. At 24 weeks, patients on placebo were re-randomized to active treatment across both dosing groups. The primary endpoint of the study evaluated the clinical effect of barzolvolimab in reducing urticaria activity (weekly urticaria activity score; UAS7) at Week 12. The studies were designed to detect a clinically meaningful difference between each of the active arms vs placebo in the overall population as well as in the subpopulation of omalizumab refractory participants. The primary endpoint analysis was performed when all patients completed the placebo controlled portion of the study at 24 weeks. A global Phase 3b long term extension study (LTE) has been established and is ongoing, which patients can enter following completion of the EMBARQ-CSU Phase 3 trials.

About Barzolvolimab
Barzolvolimab is a humanized monoclonal antibody with a novel mechanism of action that targets mast cells by binding with high specificity to a unique part of the KIT receptor and potently inhibiting its activity. The KIT receptor is abundantly expressed by mast cells and critical for their function and survival. (NYSE: AZO) today reported net sales of $6.6 billion for its fourth quarter (16 weeks) ended August 29, 2026, an increase of 5.6% from the fourth quarter of fiscal 2025 (16 weeks). Same store sales, or sales for our domestic and international stores open at least one year, are as follows:

For the quarter, gross profit, as a percentage of sales, was 53.3%, an increase of 182 basis points versus the prior year. The increase in gross margin was driven by a 145 basis point impact from tariff refunds and a 105 basis point net non-cash LIFO impact, partially offset by higher commercial mix. Operating expenses, as a percentage of sales, were 33.4% versus last year at 32.4% with deleverage primarily driven by growth initiatives.

Operating profit increased 10.1% to $1.3 billion. Net income for the quarter was $931.6 million compared to $837.0 million in the same period last year, while diluted earnings per share were $56.05 compared to last year at $48.71.

For the fiscal year ended August 29, 2026, net sales were $20.3 billion, an increase of 7.4% from the prior year. Gross profit, as a percentage of sales, was 52.3% versus last year at 52.6%. The decrease in gross margin was impacted by a 61 basis point net non-cash LIFO impact, partially offset by a 48 basis point benefit from tariff refunds. Operating expenses, as a percentage of sales, were 34.0% versus last year at 33.6%. Operating profit increased 3.1% to $3.7 billion, net income increased 3.0% to $2.6 billion and diluted earnings per share increased 5.3% to $152.55 from $144.87.

Under its share repurchase program, AutoZone repurchased 223 thousand shares of its common stock at an average price per share of $3,125, for a total investment of $697.5 million. For the fiscal year, the Company repurchased 579 thousand shares of its common stock, at an average price of $3,496, for a total investment of $2.0 billion. At year end, the Company had $1.6 billion remaining under its current share repurchase authorization.

The Company’s inventory increased 10.1% over the same period last year, driven primarily by growth initiatives. Net inventory, defined as merchandise inventories less accounts payable, on a per store basis, was negative $107 thousand versus negative $131 thousand last year and negative $107 thousand last quarter.

During the quarter ended August 29, 2026, AutoZone opened 97 new stores in the U.S., 68 in Mexico and 10 in Brazil for a total of 175 new stores. For the fiscal year, the Company opened 374 new stores. As of August 29, 2026, the Company had 6,863 stores in the U.S., 1,001 in Mexico and 167 in Brazil for a total store count of 8,031.

VANCOUVER, British Columbia, Sept. 22, 2026 (GLOBE NEWSWIRE) — Endeavour Silver Corp. (“Endeavour” or the “Company”) (NYSE: EXK; TSX: EDR) has identified a mechanical issue affecting the primary ball mill at its Guanacevi Mine.

The mill has been taken offline as of September 21, while preparations are completed to replace the mill head and trunnion, both of which are available on site. The Company expects the repairs to be completed within approximately three weeks and is coordinating with third-party service providers to expedite the replacement.

During the repair period, the regrind circuit will be configured to operate at a reduced throughput of 600 tonnes per day (tpd), compared to ordinary course capacity of approximately 1,100 tpd.

Mining operations will continue uninterrupted. The cost of replacing the affected components is expected to be minimal, as the necessary parts are already in inventory on site.
