Milestone advances Phase 3 program toward topline results expected in mid 2027

HIGH POINT, N.C., Oct. 05, 2026 (GLOBE NEWSWIRE) — vTv Therapeutics Inc. (Nasdaq: VTVT), a late-stage biopharmaceutical company advancing cadisegliatin, a novel oral investigational therapy for type 1 diabetes (T1D), today announced that the Company has completed enrollment of its Phase 3 Cadisegliatin as Adjunctive Therapy in Type 1 Diabetes (CATT1) study. A total of 166 patients have been randomized in the study evaluating cadisegliatin, a potential first-in-class oral, liver-selective glucokinase activator that has received Breakthrough Therapy designation from the U.S. Food and Drug Administration (FDA) as an adjunctive therapy to insulin for the treatment of T1D.

“Completing enrollment for the CATT1 study is a significant milestone for vTv, and we’re deeply grateful to the study participants whose commitment made this possible,” said Paul Sekhri, Chairman, President and Chief Executive Officer of vTv Therapeutics. “Cadisegliatin has the potential to reduce the risk of hypoglycemia, which remains a significant unmet need for people living with T1D, and we look forward to completing the study and reporting topline results in mid 2027.”

An estimated 9.5 million people worldwide live with T1D, and this number is projected to reach 14.7 million by 2040.i Despite advances in diabetes technology, significant gaps persist in achieving optimal day-to-day glycemic control,ii and hypoglycemia remains a daily burden on the lives of people living with T1D. The CATT1 study is evaluating the potential of cadisegliatin to reduce clinically significant hypoglycemia while improving overall glycemic control.

“This milestone is an important step forward for the cadisegliatin program,” said Thomas Strack, MD, Chief Medical Officer of vTv Therapeutics. “Our focus now is maintaining the quality and integrity of the study through completion and continuing the important work of advancing cadisegliatin development toward potential approval.”

About the Phase 3 CATT1 Study
Cadisegliatin as Adjunctive Therapy in Type 1 Diabetes (CATT1) (NCT06334133) is a randomized, double-blind, placebo-controlled Phase 3 study evaluating the efficacy and safety of cadisegliatin over six months in individuals with T1D 18 years or older who are on multiple daily insulin injections or continuous subcutaneous insulin infusion, along with continuous glucose monitoring. Participants were randomized to receive cadisegliatin 800 mg once daily, cadisegliatin 800 mg twice daily, or placebo. The primary efficacy endpoint will assess the incidence of level 2 and level 3 hypoglycemic events. Secondary endpoints include changes in hemoglobin A1c (HbA1c), time in target glucose range, and the incidence of diabetic ketoacidosis in cadisegliatin-treated participants compared with placebo. Additional information about the study is available at https://clinicaltrials.gov/study/NCT06334133.

About Cadisegliatin
Cadisegliatin (TTP399) is a novel, oral small-molecule, glucokinase activator being investigated in the U.S. as a potential first-in-class adjunctive treatment for T1D. In nonclinical studies, cadisegliatin acted selectively on the liver and increased the activity of glucokinase independently of insulin. These studies support clinical investigation of whether cadisegliatin can improve glycemic control through hepatic glucose uptake and glycogen storage. Cadisegliatin has been granted Breakthrough Therapy designation by the U.S. Food and Drug Administration (FDA).

Cadisegliatin is under investigation, and its safety and efficacy have not been established. There is no guarantee that this product will receive health authority approval or become commercially available for the use being investigated.

~ Acceptance into ETP enables early FDA engagement to support development of the RaniPill® platform technology ~

SAN JOSE, Calif., Oct. 05, 2026 (GLOBE NEWSWIRE) — Rani Therapeutics Holdings, Inc. (Nasdaq: RANI), a clinical-stage biotherapeutics company focused on oral delivery of biologics, today announced that the RaniPill® Capsule™ platform technology has been accepted into the U.S. Food and Drug Administration (FDA) Emerging Technology Program (ETP).

The FDA’s Emerging Technology Program, administered within the Agency’s Center for Drug Evaluation and Research (CDER), is designed to support the development and implementation of innovative approaches to pharmaceutical product design and manufacturing. Technologies accepted into the program must demonstrate the potential to improve pharmaceutical product quality and incorporate novel elements for which the FDA has limited review or inspection experience. Participation provides Rani with early, direct engagement with the FDA’s Emerging Technology Team to address technical and regulatory considerations related to the RaniPill platform, including through pre-submission meetings and potential site visits. Importantly, these interactions can address the broader applicability of the underlying technology rather than being limited to individual drug products.

“Our acceptance into the FDA’s Emerging Technology Program reflects the Agency’s recognition of the RaniPill® platform as a novel and innovative approach to the oral delivery of biologics,” said Talat Imran, Chief Executive Officer of Rani Therapeutics. “This program enables direct, collaborative access to the FDA on a technology as novel as Rani’s, and we expect that engagement to be especially valuable as we work to automate our vertically integrated manufacturing process in anticipation of broader clinical use. Combined with our rare disease, immunology, and obesity focused partnerships with Chugai, ProGen and PegBio, this milestone reflects our continued confidence in the RaniPill platform’s potential as an alternative to injectable biologics. We look forward to working closely with the FDA as we continue to advance the platform.”

About Rani Therapeutics
Rani Therapeutics is a clinical-stage biotherapeutics company focused on advancing technologies to enable the development of orally administered biologics and drugs. Rani has developed the RaniPill® Capsule™, a novel, proprietary and patented platform technology, intended to replace subcutaneous injection or intravenous infusion of biologics and drugs with oral dosing. Rani has successfully conducted several preclinical and clinical studies to evaluate safety, tolerability and bioavailability using RaniPill® Capsule technology.

Continued purchases and organic accumulation drive approximately 11% growth in SOL holdings since the Company’s August 12 Earnings Update

Preliminary September 30 estimates indicate double-digit SPS growth and NAV per share growth of more than 100%, both measured from August 12

In Q3, SOL outperformed the Nasdaq-100 by 56%, and DFDV outperformed SOL by 1.5x over that same period

CHAD brings Digital Credit 2.0 to income investors: a 13% annual dividend rate supported by a productive SOL treasury

BOCA RATON, FL, Oct. 05, 2026 (GLOBE NEWSWIRE) — DeFi Development Corp. (Nasdaq: DFDV) (the “Company” or “DeFi Dev Corp.”), the first U.S. public company with a treasury strategy built to accumulate and compound Solana (“SOL”), today announced that it has added approximately 26,203 SOL to its treasury since September 28, 2026, bringing total holdings to approximately 2,564,212 SOL and SOL equivalents worth $302M.

The latest increase brings total SOL and SOL equivalents growth to approximately 11% since the Company’s Q2 earnings report on August 12. With the recently established $300 million CHAD ATM providing an additional source of growth capital, the Company believes its SOL accumulation flywheel is continuing to gain momentum.

Preliminary Q3 Estimates: Per-Share Growth Metrics and Stronger Liquidity
Alongside continued accumulation, the Company expects to report substantial growth in its per-share metrics and a stronger balance sheet as of September 30, 2026. Based on preliminary estimates, the Company expects to report the following changes as of September 30, 2026, compared with August 12, 2026:

  • Double-digit growth in SOL per share (“SPS”).
  • More than 100% growth in net asset value (“NAV”) per share.
  • Double-digit growth in total SOL and SOL equivalents.
  • A reduction in notional SOL-denominated borrowings.
  • More than 100% growth in cash and cash equivalents.

These preliminary estimates reflect roughly seven weeks of rapid progress in expanding the Company’s SOL treasury, increasing SOL exposure per common share, and strengthening liquidity.

“The DFDV ship is flying at lightning speed. We have continued buying SOL and have grown our treasury by 11% since August 12,” said Joseph Onorati, Chief Executive Officer of DeFi Development Corp. “Those gains are adding up. We expect to report double digit growth in SOL per share and more than a doubling of NAV per share since our August update. Our focus remains on making each common share represent more SOL over time.”

The Company intends to continue accumulating SOL through disciplined capital deployment and organic treasury growth, while putting its holdings to work through staking and validator operations. Its capital allocation strategy remains focused on accretive growth for common shareholders alongside prudent management of liabilities and liquidity.

The anticipated September 30 metrics are preliminary, remain subject to finalization, and may differ from the Company’s final reported results. See the forward-looking statements and supplemental metric disclosures at the end of this press release.

CHAD: First Dividend Marks a Milestone in Digital Credit 2.0
On October 1, 2026, DFDV paid CHAD’s first dividend, marking a milestone for its Nasdaq-listed variable-rate preferred stock. CHAD currently carries a 13% annual dividend rate on its $10 stated amount, equivalent to $1.30 per share annually at the current rate, with dividend payments continuing each business day when declared.

With payments underway, the Company is focused on expanding investor awareness, market access, and liquidity as it works toward establishing CHAD at its $10 stated amount and scaling it as a source of capital for DFDV’s next phase of growth.

The Company views CHAD as Digital Credit 2.0: preferred equity supported by a productive treasury. Through staking and validator operations, DFDV’s SOL holdings generate recurring rewards and fees, providing an organic source of income to support dividend obligations. The Company continues to maintain ample dividend coverage, while additional SOL accumulation can expand the treasury’s earning capacity over time.

“CHAD is fuel for the DFDV engine and central to our next phase of growth,” said Joseph Onorati, Chief Executive Officer of DeFi Development Corp. “It opens our strategy to income investors and gives us another source of capital to accumulate SOL and expand the treasury’s earning power. Our focus now is on building investor awareness, deepening liquidity, and establishing CHAD at its $10 stated amount.”

“We see a significant opportunity to scale CHAD from here,” continued Onorati. “Trading at par would position us to raise additional perpetual capital on terms that support accretive growth for common shareholders, while offering CHAD holders a compelling income stream. That is the next phase we are working toward.”

Forward Looking Statements and Metrics
This press release contains “forward-looking statements” within the meaning of the safe harbor provisions of the U.S. Private Securities Litigation Reform Act of 1995. Forward-looking statements in this press release include, but are not limited to, statements regarding expected September 30, 2026 metrics and growth in SPS, NAV per share, SOL holdings and cash balances; anticipated changes in borrowings and liquidity; future SOL accumulation and per-share accretion; capital raising and deployment, the anticipated issuance price of shares under the CHAD ATM program, the CHAD dividend rate, the declaration and payment of CHAD dividends, the intended use of proceeds, and the Company’s ability to expand its SOL treasury and increase revenue, and can be identified by words such as “anticipate,” “intend,” “plan,” “believe,” “project,” “estimate,” “expect,” “strategy,” “future,” “likely,” “may,” “should,” “will” and similar references to future periods. Forward-looking statements are neither historical facts nor assurances of future performance. Instead, they are based only on the Company’s current beliefs, expectations, and assumptions regarding the future of its business, future plans and strategies, projections, anticipated events and trends, the economy, and other future conditions. Because forward-looking statements relate to the future, they are subject to inherent uncertainties, risks, and changes in circumstances that are difficult to predict, many of which are outside of the Company’s control. The Company’s actual results and financial condition may differ materially from those indicated in the forward-looking statements. Therefore, you should not rely on any of these forward-looking statements.

Important factors that could cause our actual results and financial condition to differ materially from those indicated in the forward-looking statements include, among others, the following: (i) fluctuations in the market price of SOL and any associated losses that the Company may incur as a result of a decrease in the market price of SOL; (ii) a failure for the demand for SOL, or activity on the SOL network, to continue to develop and grow we anticipate or at all; (iii) volatility in our stock price, including due to future issuances of common stock and securities convertible into common stock; (iv) the effect of and uncertainties related to the ongoing volatility in interest rates; (v) our ability to achieve and maintain profitability in the future; (vi) the impact on our business of the regulatory environment and complexities of complying with such environment, including changes in securities laws or other laws or regulations; (vii) changes in the accounting treatment relating to the Company’s SOL holdings; (viii) our ability to respond to general economic conditions; (ix) our ability to manage our growth effectively and our expectations regarding the development and expansion of our business; (x) our ability to access sources of capital, including debt financing and other sources of capital to finance operations and growth; and (xi) the risk that the Company’s preliminary estimates as of September 30, 2026 differ materially from its final results; (xii) risks relating to CHAD, including that the dividend rate may be adjusted, that dividends are payable only when declared, that CHAD may trade below its stated amount and that staking and validator rewards may be insufficient to support dividend payments; (xiii) other risks and uncertainties more fully described in the section captioned “Risk Factors” in the Company’s most recent Annual Report on Form 10-K and other reports we file with the SEC.

As a result of these matters, changes in facts, assumptions not being realized, or other circumstances, the Company’s actual results may differ materially from the expected results discussed in the forward-looking statements contained in this press release. Forward-looking statements contained in this announcement are made as of this date, and the Company undertakes no duty to update such information except as required under applicable law.

SPS and NAV per share are supplemental analytical measures used by management to assess the Company’s treasury strategy and economic exposure attributable to common shareholders. For purposes of this release, NAV reflects the value of SOL and SOL equivalents, plus cash and cash equivalents, less SOL-denominated liabilities, outstanding principal of out-of-the-money convertible debt, other debt, and the aggregate notional amount of preferred equity. NAV per share reflects that amount divided by adjusted common shares outstanding, which includes ordinary shares outstanding, shares underlying restricted stock units and vested options, and applicable dilution from assumed exercise of in-the-money warrants and conversion of in-the-money convertible securities. Related adjustments to NAV reflect assumed warrant exercise proceeds and the removal of debt assumed converted, as applicable, to avoid double counting. SPS expresses the corresponding net SOL exposure per adjusted common share, with the SPS growth comparison applying the updated, liability-adjusted methodology and a SOL reference price to both measurement dates. These measures depend on valuation and capital structure assumptions and do not represent GAAP book value per share, liquidation proceeds, or shareholder investment returns. Preliminary estimates are not audited and remain subject to completion of the Company’s financial close and verification of underlying balances and share counts, and final results may differ materially.

To learn more about Pacira, visit www.pacira.com.

About EXPAREL® (bupivacaine liposome injectable suspension)

EXPAREL is indicated to produce postsurgical local analgesia via infiltration in patients aged 6 years and older, and postsurgical regional analgesia via an interscalene brachial plexus block in adults, a sciatic nerve block in the popliteal fossa in adults, and an adductor canal block in adults. The safety and effectiveness of EXPAREL have not been established to produce postsurgical regional analgesia via other nerve blocks besides an interscalene brachial plexus nerve block, a sciatic nerve block in the popliteal fossa, or an adductor canal block. The product combines bupivacaine with multivesicular liposomes, a proven product delivery technology that delivers medication over a desired time period. EXPAREL represents the first and only multivesicular liposome local anesthetic that can be utilized in the peri- or postsurgical setting. By utilizing the multivesicular liposome platform, a single dose of EXPAREL delivers bupivacaine over time, providing significant reductions in cumulative pain scores with up to a 78 percent decrease in opioid consumption; the clinical benefit of the opioid reduction was not demonstrated. Additional information is available at www.EXPAREL.com.

Important Safety Information about EXPAREL for Patients

EXPAREL should not be used in obstetrical paracervical block anesthesia. In studies in adults where EXPAREL was injected into a wound, the most common side effects were nausea, constipation, and vomiting. In studies in adults where EXPAREL was injected near a nerve, the most common side effects were nausea, fever, and constipation. In the study where EXPAREL was given to children, the most common side effects were nausea, vomiting, constipation, low blood pressure, low number of red blood cells, muscle twitching, blurred vision, itching, and rapid heartbeat. EXPAREL can cause a temporary loss of feeling and/or loss of muscle movement. How much and how long the loss of feeling and/or muscle movement depends on where and how much of EXPAREL was injected and may last for up to 5 days. EXPAREL is not recommended to be used in patients younger than 6 years old for injection into the wound, for patients younger than 18 years old, for injection near a nerve, and/or in pregnant women. Tell your health care provider if you or your child has liver disease, since this may affect how the active ingredient (bupivacaine) in EXPAREL is eliminated from the body. EXPAREL should not be injected into the spine, joints, or veins. The active ingredient in EXPAREL can affect the nervous system and the cardiovascular system; may cause an allergic reaction; may cause damage if injected into the joints; and can cause a rare blood disorder.

Trial follows promising MAR002 Phase 1 clinical trial results that demonstrated deep and durable IGF-1 suppression with PK properties that may enable dosing as infrequently as once every two weeks, compared to the daily subcutaneous injections required by the current standard of care

By directly blocking growth hormone signaling at the receptor level, MAR002 has the potential to deliver meaningful biochemical control for a broad population of acromegaly patients

SOUTH SAN FRANCISCO, Calif. and LIBERTY CORNER, N.J., Oct. 05, 2026 (GLOBE NEWSWIRE) — Marea Therapeutics, LLC (“Marea”), a subsidiary of Lisata Therapeutics, Inc. (Nasdaq: LSTA) (“Lisata”), a clinical-stage biotechnology company harnessing the latest advances in human genetics to develop first-in-class, next-generation medicines for cardioendocrine diseases, today announced the first patient has been dosed in its Phase 2 clinical trial of MAR002 for the treatment of acromegaly.

Acromegaly is a rare, chronic, progressive endocrine disorder caused by prolonged exposure to excess growth hormone (GH), most commonly due to a GH-secreting pituitary adenoma, resulting in elevated insulin-like growth factor-1 (IGF-1). It leads to the abnormal enlargement of the hands, feet, and facial features, and if left untreated, can result in severe organ damage, systemic complications, and early mortality.

MAR002 is a first-in-class allosteric monoclonal antibody investigational therapy targeting the growth hormone receptor (GHR). Data from a first-in-human Phase 1 study with MAR002 presented at the 2026 Annual Meeting of the Endocrine Society (ENDO) in June 2026 demonstrated compelling proof-of-mechanism with deep and durable IGF-1 suppression and pharmacokinetic (PK) properties that may enable dosing as infrequently as once every two weeks, compared to the daily subcutaneous injections required by the current standard of care.

“Treating our first acromegaly patient with MAR002 represents an important milestone for Marea Therapeutics,” said Josh Lehrer, M.D., president and chief operating officer of Lisata Therapeutics and chief executive officer of Marea Therapeutics. “We believe MAR002 has a potential best-in-class profile and, if approved, could be transformative for the majority of acromegaly patients who cannot achieve disease control with current therapies. We are focused on clinical execution to rapidly advance this product candidate to acromegaly patients who need new treatment options.”

“People living with acromegaly often face years of symptoms before receiving a diagnosis, and even after diagnosis, many continue to struggle with the long-term burden of managing this chronic disease,” said Jill Sisco, president of Acromegaly Community, Inc. “The advancement of new treatment options that have the potential to provide effective disease control while reducing treatment burden offers hope for the acromegaly community. We are encouraged to see continued investment in investigational therapies like MAR002 that aim to address the needs and priorities of people living with this condition and expand treatment options.”

This study intends to enroll approximately 72 participants with acromegaly across three MAR002 dose arms, after an initial 900 mg loading dose, compared to placebo, with a primary endpoint of IGF-1 reduction following 10 weeks of treatment. After completing the 10-week treatment period, eligible participants will have an option to enroll in an open-label extension clinical trial.

About Acromegaly
Acromegaly is an orphan disease characterized by the excess secretion of growth hormone (GH) from a benign pituitary adenoma. Acromegaly affects approximately 30,000 patients in the U.S. If left untreated, acromegaly is highly morbid, leading to significant comorbidities such as GH-induced insulin resistance and diabetes, and serious cardiovascular pathology. The median lifespan of patients can be shortened by 10 years without effective therapy, and incomplete IGF-1 normalization is associated with increased mortality. Despite its severity, acromegaly is often under or misdiagnosed, with an average time from symptom onset to diagnosis of approximately eight years.

About MAR002
MAR002 is a potent and selective half-life-extended, allosteric, human monoclonal growth hormone receptor antagonist (GHRA) antibody being developed for the treatment of acromegaly. The in vivo PK and PD properties of MAR002 have been observed to be predictable and typical of a half-life extended human antibody, showing a long duration of action compatible with infrequent subcutaneous dose administration in humans. These characteristics support its potential to offer an effective and convenient treatment for patients with acromegaly, if approved.

New collaboration combines Zscaler Autonomous Application Shield with Lightwell from IBM and Red Hat to help organizations reduce exposure between vulnerability disclosure and patch deployment through inline protection and validated remediation

SAN JOSE, Calif., Oct. 05, 2026 (GLOBE NEWSWIRE) — Zscaler, Inc. (NASDAQ: ZS), the cybersecurity platform for the AI era, today announced a collaboration with IBM and Red Hat to help organizations protect private applications during the period between vulnerability disclosure and patch deployment. The collaboration combines Zscaler Autonomous Application Shield, delivered through the Zscaler Zero Trust ExchangeTM security platform, with Lightwell from IBM and Red Hat to deliver adaptive, application-specific protection with validated software remediation in a single coordinated response.

The economics of exploitation have shifted. Capabilities that once required elite skills and weeks of manual effort are now widely available, and attackers are weaponizing newly disclosed vulnerabilities faster than enterprise patch cycles can respond. According to Mandiant’s M-Trends 2026 report, the mean time to exploit a vulnerability has fallen below zero, meaning vulnerabilities are increasingly exploited before a public patch exists. Patching remains necessary, but on its own it can no longer keep pace. Organizations need protection that can be applied at machine speed while remediation is prepared and deployed.

This collaboration extends Zscaler’s protection-first operating model for private applications, which assumes a remediation window will always exist and builds resilience around it: hide applications with Zscaler Private AccessTM (ZPATM) so they are harder to discover and target, segment access with Autonomous User-to-App Segmentation so a foothold does not become broad reachability, and shield reachable applications with Autonomous Application Shield so exploit attempts are stopped inline while the root cause is fixed. By uniting Zscaler Autonomous App Shield with Lightwell, the collaboration provides customers a coordinated path from immediate protection to durable fix.

Operating through the Zero Trust Exchange, Zscaler App Connectors continuously perform safe assessments of each private application’s exposure points and vulnerabilities. The Zscaler cloud individually evaluates each application, ensuring only necessary protections are applied. When a new vulnerability emerges, adaptive protections can be applied inline and in real time, without waiting for a patch. Through this collaboration, Lightwell helps Zscaler defend its customers from potential cyber threats, with Zscaler blocking exploit attempts immediately at the network level. Lightwell then provides validated remediations for affected open source software, enabling customers to fix the underlying flaw at the source.

“As attackers weaponize newly disclosed vulnerabilities faster than enterprises can respond, organizations need a way to protect private applications immediately while fixes are being prepared and deployed,” said Joby Menon, Senior Vice President of Product Management at Zscaler. “Our collaboration with IBM and Red Hat brings together inline, application-specific protection and validated remediation to help customers reduce risk during that gap.”

Through this collaboration, Zscaler, IBM, and Red Hat will deliver integrated capabilities across the application security lifecycle, addressing critical needs including:

  • Exposure Discovery: Providing a current understanding of a private application’s actual risk profile, updated as fast as applications change, rather than relying on periodic scans.
  • Preemptive, Application-Specific Protection: Bridging the gap between vulnerability and patching. Applying only the protections a given application needs during the gap between disclosure and patching, reducing exposure while remediation is underway and avoiding one-size-fits-all policy noise.
  • Machine-Speed Response: Deploying protections inline and in real time when a new vulnerability emerges, informed by global threat intelligence from across Zscaler’s worldwide customer base.
  • Validated Source-Level Remediation: Delivering tested software fixes through Lightwell so organizations can resolve the underlying vulnerability, with advisory and deployment support to prioritize and validate protections across complex environments.

“AI is shrinking the window between vulnerability discovery and vulnerability exploitation. A window that was months or weeks is now hours and minutes,” said Gunnar Hellekson, vice president and general manager, Lightwell, Red Hat. “Lightwell helps keep open source safe by patching and fixing these vulnerabilities at machine speed. Collaborating with partners like Zscaler brings Lightwell’s capabilities to the forefront of enterprise networks, mitigating potential vulnerabilities head-on while patches are applied to critical systems.”

Autonomous Application Shield is currently available through an Early Access Program. Availability, capabilities, and timing of the collaboration described herein are subject to change.
