STAMPEDE study analysis providing new predictive evidence in clinically very high-risk, nonmetastatic disease to be presented at ESMO
SOUTH SAN FRANCISCO, Calif., Oct. 23, 2026 (GLOBE NEWSWIRE) — Veracyte, Inc. (Nasdaq: VCYT), a leading cancer diagnostics company, today announced new data showing that its Decipher® Prostate Genomic Classifier can predict benefit from adding abiraterone to androgen deprivation therapy (ADT) in men with clinically very high-risk nonmetastatic prostate cancer. The findings will be presented tomorrow in a proffered paper oral presentation at the European Society for Medical Oncology (ESMO) Congress 2026 by Dr. Sarah Howlett of University College London (UCL).
The retrospective analysis of the randomized Phase III STAMPEDE trial provides new Level 1B predictive evidence for Decipher in men with clinically very high-risk nonmetastatic prostate cancer. This predictive evidence bolsters Decipher’s high-risk claims to help physicians identify which patients are most likely to benefit from adding abiraterone to ADT. These findings build on Decipher’s long-established standing in guidelines for assessing the risk of cancer progression, as well as prior STAMPEDE analysis that was recently incorporated into the NCCN 2027 prostate cancer guidelines for high- and very high-risk disease.
For patients with very high-risk disease, the decision to add abiraterone treatment carries significant consequences. Abiraterone can improve cancer outcomes when added to ADT but also adds side effects and treatment burden. Understanding a patient’s expected benefit from the additional treatment can help physicians and patients weigh the potential benefit alongside the impact on daily life.
“For men with very high-risk disease, preventing the cancer from spreading is critical, and understanding who benefits most from additional treatment can make a meaningful difference,” said Dr. Sarah Howlett, clinical research fellow in the Research Department of Oncology at UCL and the study’s presenter. “This analysis shows that the Decipher test can identify which patients in this study population were at greatest risk of prostate cancer-specific mortality and, therefore, may gain the greatest benefit from adding abiraterone to ADT. That gives physicians and patients evidence to make more confident treatment decisions based on the biology of the individual tumor.”
Key study findings in very high-risk patients:
The STAMPEDE analysis included 401 patients who received ADT alone or ADT plus abiraterone. The separate real-world analysis evaluated associations between Decipher scores and patient outcomes; it was not designed to assess prediction of abiraterone benefit.
“These findings expand Decipher’s predictive evidence to another consequential treatment decision: whether adding abiraterone will provide meaningful benefit for a man with very high-risk prostate cancer,” said Elai Davicioni, Ph.D., Veracyte’s medical director, Urology. “Building on the predictive data recently presented in intermediate-risk disease at ASTRO, these results strengthen Decipher’s role in helping physicians connect tumor biology to treatment selection across the prostate cancer risk continuum. We believe this growing evidence gives physicians a stronger reason to use Decipher before making decisions that can affect both cancer outcomes and patients’ quality of life.”
Decipher is already used in the care of patients with high-risk prostate cancer. The new findings strengthen the clinical rationale for testing before treatment-intensification decisions and support the potential for broader adoption and use in this population. Another trial prospectively evaluating the use of the Decipher test to guide treatment intensification or de-intensification in high-risk prostate cancer is the phase III biomarker-driven NRG-GU009 (PREDICT-RT) study, which completed enrollment earlier this year. This trial will add prospective evidence to the findings from the STAMPEDE trial, further expanding the predictive evidence around the Decipher test for treatment intensification with apalutamide, a different androgen receptor pathway inhibitor.

In a 1L EGFR atypical population with more than double the rate of baseline CNS metastases as competitor datasets, enozertinib demonstrates a potential best-in-class 84% confirmed systemic ORR and 70% confirmed intracranial ORR, with multiple intracranial complete responses
88% 6-month landmark PFS compares favorably to current standards of care and to investigational therapies; follow-up ongoing with majority of patients continuing on treatment
Competitive safety profile with no significant off-target toxicity and no treatment-related discontinuations
Redwood-1 global Phase 3 registrational trial initiation in 1L EGFR atypical NSCLC expected in 1H 2027
Company to host a conference call and webcast today at 8:00 am ET featuring Dr. Joshua Sabari
SOUTH SAN FRANCISCO, Calif. and SAN DIEGO, Oct. 23, 2026 (GLOBE NEWSWIRE) — ORIC Pharmaceuticals, Inc. (Nasdaq: ORIC), a clinical stage oncology company focused on developing and commercializing treatments that address mechanisms of therapeutic resistance, today presented potential best-in-class data for enozertinib in first-line NSCLC patients with EGFR atypical mutations at ESMO Congress 2026 and announced plans to advance enozertinib into a global Phase 3 trial.
“These enozertinib systemic and intracranial response data set a new benchmark in 1L EGFR atypical NSCLC,” said Pratik S. Multani, M.D., chief medical officer. “The data are particularly impressive given that nearly two-thirds of our patients had baseline brain metastases, which is more than double the rate seen in competitor datasets, and which has historically been associated with poorer outcomes.”
“Supported by these potential best-in-class data and having successfully completed End-of-Phase 1 interactions with FDA, we are rapidly advancing enozertinib into a global Phase 3 registrational trial,” said Jacob M. Chacko, M.D., president and chief executive officer. “With rinzimetostat’s first Phase 3 trial in prostate cancer initiated earlier this year and enozertinib’s first Phase 3 trial in lung cancer planned for 1H 2027, ORIC has become a diversified, late-stage oncology company with two programs offering opportunities for broader development across indications with significant unmet need and meaningful commercial potential.”
Enozertinib Phase 1b Data in 1L EGFR Atypical NSCLC
Enozertinib is being evaluated in an ongoing Phase 1b trial in patients with locally advanced or metastatic NSCLC with EGFR atypical and EGFR exon 20 insertion mutations. Notably, enrollment allows patients with previously untreated brain metastases (i.e., active, untreated brain metastases). Key objectives of the trial included safety and determination of the recommended Phase 2 dose, along with assessment of antitumor activity based on investigator-assessed objective response rate (ORR), blinded independent central review (BICR)-assessed ORR, CNS-ORR according to modified RECIST 1.1 and/or Response Assessment in Neuro-Oncology Brain Metastases (RANO-BM), as well as DOR and PFS.
As of the September 4, 2026 cutoff date, data are presented for 26 patients who were treated with 80 mg once daily oral enozertinib, including 23 patients with P-loop and alpha C-helix compressing (PACC) mutations and 3 patients with L861Q mutations without a co-occurring PACC mutation. Brain metastases were present in 65% of patients at baseline, which is more than double the rate versus competitor datasets. Notably, 46% of all patients had previously untreated brain metastases.
Safety Analysis
Enozertinib was well tolerated with predominantly Grade 1 or 2 treatment-related adverse events (TRAEs) and no significant off-target toxicities. Most frequent TRAEs included diarrhea, paronychia, and dermatitis acneiform. Six patients (23%) experienced Grade 3 TRAEs, all of which were resolved with dose modifications. There were no Grade 4 or 5 TRAEs and no treatment discontinuations related to TRAEs.
Meaningful Systemic Antitumor Activity
Tumor reductions were observed across all response-evaluable patients1. Reductions were also observed across a wide range of mutations, including the most prevalent PACC mutations (e.g., G719X, S768I), a broad spectrum of PACC complex mutations, and L861Q mutations. Enozertinib demonstrated meaningful systemic activity, including:
Deep EGFR-mutant ctDNA molecular responses were achieved by Cycle 3 Day 1 in all response-evaluable patients across diverse mutations2:
Compelling Intracranial Antitumor Activity
Enozertinib continues to demonstrate potential best-in-class intracranial efficacy. Notably, no new CNS lesions were observed in patients with or without baseline CNS metastases.
All response-evaluable patients with measurable CNS disease (90% without prior treatment for their CNS metastases) achieved CNS tumor reductions per investigator-assessment3. Enozertinib demonstrated compelling intracranial activity, including:
Intracranial efficacy was also evaluated utilizing the more stringent BICR RANO-BM assessment criteria. Based on this assessment, CNS tumor reductions were observed in nearly all response-evaluable patients, including in patients with previously untreated CNS metastases4. Enozertinib demonstrated compelling intracranial activity, including:
Promising Early Durability
Median progression-free survival and median duration of response data are immature. The Kaplan-Meier 6-month landmark PFS is 88%. Follow-up is ongoing with the majority of patients continuing on treatment.
Redwood-1 Phase 3 Initiation
Supported by potential best-in-class data presented at ESMO Congress 2026 and based on End-of-Phase 1 interactions with the FDA, ORIC plans to initiate Redwood-1, a global Phase 3 registrational trial in EGFR atypical NSCLC in 1H 2027. This trial plans to enroll approximately 360 patients with PACC and L861X mutations, including those with untreated brain metastases, randomized to receive 80 mg once daily oral enozertinib or physician’s choice of osimertinib or afatinib. Dual primary endpoints include ORR by BICR for potential accelerated approval as well as PFS by BICR for full approval. Key secondary endpoints include overall survival and intracranial ORR by BICR RANO-BM.

Preclinical data highlight potential advantages of targeting PRC2 via EED over EZH2, including the emergence of resistance mutations to EZH2 inhibitors like mevrometostat and against which the EED inhibitor rinzimetostat maintained effectiveness
Preclinical results, supported by translational data in mCRPC patient samples, show rinzimetostat reinforces a luminal, AR-dependent state, restricts lineage plasticity and enhances sensitivity to AR inhibition
Findings reinforce rinzimetostat’s differentiated profile and support development across the prostate cancer disease spectrum, including in the ongoing Himalayas-1 Phase 3 trial
SOUTH SAN FRANCISCO, Calif. and SAN DIEGO, Oct. 23, 2026 (GLOBE NEWSWIRE) — ORIC Pharmaceuticals, Inc. (Nasdaq: ORIC), a clinical stage oncology company focused on developing and commercializing treatments that address mechanisms of therapeutic resistance, today announced preclinical and translational data from an ePoster presentation at ESMO Congress 2026 highlighting the potential of rinzimetostat to enhance sensitivity to androgen receptor inhibition in prostate cancer.
“These results strengthen the rationale for combining PRC2 inhibition with androgen receptor targeted therapies across castration-sensitive and castration-resistant prostate cancer by limiting tumor adaptability and enhancing sensitivity to AR inhibition,” said Lori Friedman, PhD, chief scientific officer. “Verification of our preclinical results in patient samples, together with the potential advantages of targeting PRC2 via EED, reinforce our confidence in rinzimetostat’s potential best-in-class profile and support its evaluation across the prostate cancer disease spectrum, including in our ongoing Himalayas-1 Phase 3 trial.”
Rinzimetostat, a Next Generation PRC2 Inhibitor, Enhances Luminal Fate and AR Signaling While Restricting Lineage Plasticity in Preclinical Models and Demonstrates Mechanistic Proof-of-Concept in Samples from Patients with mCRPC
ePoster Number: 1128eP
Session Title: Developmental Therapeutics

SHANGHAI, China, Oct. 23, 2026 (GLOBE NEWSWIRE) — On October 21, Professor Joseph Sifakis, a Turing Award laureate and Chief Scientific Advisor of the CaoCao Mobility AI Innovation Research Center, visited the center for in-depth discussions with CaoCao’s technology team. On the same day, CaoCao launched a research project on Trustworthy Decision-Making and Multi-Agent Coordination for Robotaxi Curbside Operations. Professor Sifakis witnessed the launch and offered valuable recommendations based on his extensive research experience.

Professor Sifakis is one of the pioneers of model checking. He received the 2007 Turing Award for his contributions to the development of model-checking theory and its applications. He has long been engaged in research on complex intelligent systems and Trustworthy Autonomous Systems, areas closely related to the safety and trustworthiness challenges facing Robotaxi operations at scale.
In June this year, Professor Sifakis was formally invited to serve as Chief Scientific Advisor of the CaoCao Mobility AI Innovation Research Center to provide academic guidance for CaoCao’s AI transformation and the development of its autonomous driving technologies. At the 2026 World Artificial Intelligence Conference, he outlined three fundamental principles for trustworthy Robotaxis: Safe by Design, System Coordination, and Runtime Assurance. He also noted that CaoCao’s RoboX ecosystem closely aligns with the systems-engineering principles underlying Trustworthy Autonomous Systems.
Passenger pick-up and drop-off are critical to the Robotaxi experience. This newly launched research project focuses on safety and trustworthiness in this everyday mobility scenario, exploring how the theory of Trustworthy Autonomous Systems can be translated into verifiable engineering methods.
Drawing on 11 years of real-world operating data and experience from CaoCao’s mobility services, the project will examine the conditions under which curbside stopping locations can be used, passenger boarding and alighting paths, vehicle stopping behavior, and system coordination. By exploring methods for cross-validating results from simulation and real-vehicle testing, the project aims to build a body of reusable, scenario-based evidence to support safety validation for the trustworthy transfer of Robotaxi operations across cities and their deployment worldwide.
