In addition to the above poster presentations, Ardelyx will host an Exhibitor Spotlight session titled “Expanding Perspectives in Hyperphosphatemia Treatment: A Different Approach” on Friday, October 23, 2026, at 11:00 am MDT. Leading nephrologist Samuel A. Kantor, MD, will present an in-depth look at XPHOZAH (tenapanor), a first-in-class phosphate absorption inhibitor (PAI). Dr. Kantor will cover XPHOZAH’s mechanism of action, efficacy and safety data, and practical guidance for its use as an add-on therapy for dialysis patients who remain above guideline-established serum phosphorus targets despite current phosphate binder treatment.

About XPHOZAH® (tenapanor)
XPHOZAH® (tenapanor) is the first and only phosphate absorption inhibitor (PAI) approved by the U.S. Food and Drug Administration to reduce serum phosphorus in adults with chronic kidney disease (CKD) on dialysis as add-on therapy in patients who have an inadequate response to phosphate binders or who are intolerant of any dose of phosphate binder therapy. With a differentiated mechanism of action, XPHOZAH blocks phosphate absorption through the paracellular pathway, the primary pathway of phosphate absorption, and is administered as one tablet taken twice daily. Diarrhea was the most common side effect experienced by patients taking XPHOZAH in clinical trials. Please see Important Safety Information below and click for full Prescribing Information.

About Hyperphosphatemia
Hyperphosphatemia is a serious condition characterized by elevated levels of phosphate in the blood that affects many of the approximately 550,000 people in the United States with chronic kidney disease (CKD) on maintenance dialysis. As kidney function declines, the kidneys are unable to adequately eliminate excess phosphate, leading to its accumulation in the blood. Elevated phosphate levels are associated with adverse clinical outcomes in patients with CKD on dialysis. International KDIGO treatment guidelines recommend lowering progressively or persistently elevated serum phosphate toward the laboratory-specific normal range. In adults, a commonly used reference range is approximately 2.5–4.5 mg/dL.

IMPORTANT SAFETY INFORMATION
CONTRAINDICATIONS
XPHOZAH is contraindicated in:

  • Pediatric patients under 6 years of age
  • Patients with known or suspected mechanical gastrointestinal obstruction

WARNINGS AND PRECAUTIONS
Diarrhea
Patients may experience severe diarrhea. Treatment with XPHOZAH should be discontinued in patients who develop severe diarrhea.

MOST COMMON ADVERSE REACTIONS
Diarrhea, which occurred in 43-53% of patients, was the only adverse reaction reported in at least 5% of XPHOZAH-treated patients with CKD on dialysis across trials. The majority of diarrhea events in the XPHOZAH-treated patients were reported to be mild-to-moderate in severity and resolved over time, or with dose reduction. Diarrhea was typically reported soon after initiation but could occur at any time during treatment with XPHOZAH. Severe diarrhea was reported in 5% of XPHOZAH-treated patients in these trials.

INDICATION
XPHOZAH (tenapanor), 30 mg BID, is indicated to reduce serum phosphorus in adults with chronic kidney disease (CKD) on dialysis as add-on therapy in patients who have an inadequate response to phosphate binders or who are intolerant of any dose of phosphate binder therapy.

For additional safety information, please see full Prescribing Information.

— Global, first-in-human study underway designed to evaluate safety, tolerability, and initial clinical efficacy of PM577a —

America and Europe‌ —

— PM577 granted Rare Pediatric Disease Designation by the U.S. FDA —

— Initial clinical data expected in 2027 —

CAMBRIDGE, Mass., Oct. 05, 2026 (GLOBE NEWSWIRE) — Prime Medicine, Inc. (Nasdaq: PRME), a biotechnology company committed to delivering a new class of differentiated one-time curative genetic therapies, today announced that the first patient has been dosed in the Company’s global Phase 1/2 clinical trial of PM577a, an investigational in vivo Prime Editor for H1069Q-mutated Wilson disease. The milestone marks the initiation of the first clinical study of an in vivo Prime Editing therapy from Prime Medicine, as well as the first clinical evaluation of the company’s liver franchise. The company also announced that the U.S. Food and Drug Administration (FDA) has granted Rare Pediatric Disease (RPD) designation to PM577 for Wilson disease.

“Dosing the first patient with PM577a is a meaningful step toward our goal of delivering a transformative, one-time treatment for people living with Wilson disease,” said Allan Reine, M.D., Chief Executive Officer of Prime Medicine. “We’ve seen strong interest from patients and physicians that, together with our patient identification and prescreening efforts, positions us well for efficient enrollment. More broadly, PM577a reflects the modularity of our platform. While this initial candidate is designed to correct H1069Q, the most prevalent disease-causing mutation in the United States and Europe, the same shared LNP delivery approach provides a path to expand into additional Wilson disease patient populations globally, beginning with our follow-on candidate designed to correct the R778L mutation, the most common mutation in East Asian populations.”

“Wilson disease is a serious, progressive genetic disorder with no approved curative therapy. Current pharmacologic treatments require lifelong management and can be limited by tolerability and adherence challenges,” said Mohammed Asmal, M.D., Ph.D., Chief Medical Officer of Prime Medicine. “PM577a is designed to correct the H1069Q mutation at its genetic root through a single administration, with the potential to offer patients a one-time curative treatment, ideally before irreversible liver and neurological damage occur. Because Wilson disease frequently presents during childhood and adolescence, we are particularly encouraged by the FDA’s Rare Pediatric Disease designation for PM577, which underscores the significant unmet need for therapies capable of altering the course of disease early in life.”

Phase 1/2 Clinical Trial

The Phase 1/2 clinical trial is an open-label, global, first-in-human study designed to evaluate the safety, tolerability, and preliminary clinical efficacy of ascending doses of PM577a in adults and adolescents with Wilson disease containing at least one p.H1069Q allele in the ATP7B gene. The study will initially enroll adults who are clinically stable on standard-of-care therapy. Initial efficacy measures may include copper efflux by 64Cu PET, serum ceruloplasmin, non-ceruloplasmin-bound copper, and 24-hour urinary copper excretion, among others. The trial will also evaluate discontinuation of baseline standard-of-care therapies.

Additional information about the trial is available at ClinicalTrials.gov under identifier NCT07748403.

Rare Pediatric Disease Designation

FDA grants Rare Pediatric Disease designation to investigational therapies intended to treat serious or life-threatening diseases that primarily affect individuals from birth through 18 years of age and affect fewer than 200,000 people in the United States. Under the FDA’s Rare Pediatric Disease Priority Review Voucher (PRV) program, a sponsor that receives an approval for a New Drug Application (NDA) or Biologics License Application (BLA) for a rare pediatric disease may qualify for a voucher that can be redeemed to receive a priority review for a subsequent marketing application, or it can be sold or transferred to another company.

About PM577

PM577 is a family of lipid nanoparticle-formulated Prime Editing products designed to correct pathogenic ATP7B mutations in hepatocytes through a single intravenous infusion. Prime Medicine’s initial candidate, PM577a, targets the ATP7B H1069Q mutation, the most prevalent Wilson disease-causing allele in North America and Europe. PM577a has been granted Rare Pediatric Disease designation by the U.S. FDA for the treatment of Wilson disease. The Company intends to develop additional products to address the majority of pathogenic variants on a global basis. A follow-on candidate is in preclinical development targeting R778L, the most common mutant allele in East Asian populations.

About Wilson Disease

Wilson disease is a rare, serious and progressive genetic disorder of hepatic copper transport caused by loss-of-function mutations in the ATP7B gene. Impaired biliary excretion drives progressive copper accumulation in the liver, followed by multiorgan involvement, including the brain, kidneys and cornea. Current pharmacologic therapies, including copper chelators and zinc salts, are non-curative, require lifelong daily treatment and carry tolerability and adherence challenges. Liver transplantation remains the only curative option but is constrained by organ availability and carries substantial procedural risk. PM577a is designed to offer patients with H1069Q-mutated Wilson disease a one-time therapy that corrects the disease at its genetic root.

About Prime Medicine

Prime Medicine is a leading biotechnology company dedicated to creating and delivering the next generation of gene editing therapies to patients. The Company is deploying its proprietary Prime Editing platform, a versatile, precise and efficient gene editing technology, to develop a new class of differentiated one-time curative genetic therapies. Designed to make only the right edit at the right position within a gene while minimizing unwanted DNA modifications, Prime Editors have the potential to repair almost all types of genetic mutations and work in many different tissues, organs and cell types. Taken together, Prime Editing’s versatile gene editing capabilities could unlock opportunities across thousands of potential indications.

Prime Medicine is currently progressing a diversified portfolio of investigational therapeutic programs organized around its core areas of focus: liver, lung, and immunology and oncology. Across each core area, Prime Medicine is focused initially on a set of high-value programs, each targeting a disease with well-understood biology and a clearly defined clinical development and regulatory path, and each expected to provide the foundation for expansion into additional opportunities. Milestone advances Phase 3 program toward topline results expected in mid 2027

HIGH POINT, N.C., Oct. 05, 2026 (GLOBE NEWSWIRE) — vTv Therapeutics Inc. (Nasdaq: VTVT), a late-stage biopharmaceutical company advancing cadisegliatin, a novel oral investigational therapy for type 1 diabetes (T1D), today announced that the Company has completed enrollment of its Phase 3 Cadisegliatin as Adjunctive Therapy in Type 1 Diabetes (CATT1) study. A total of 166 patients have been randomized in the study evaluating cadisegliatin, a potential first-in-class oral, liver-selective glucokinase activator that has received Breakthrough Therapy designation from the U.S. Food and Drug Administration (FDA) as an adjunctive therapy to insulin for the treatment of T1D.

“Completing enrollment for the CATT1 study is a significant milestone for vTv, and we’re deeply grateful to the study participants whose commitment made this possible,” said Paul Sekhri, Chairman, President and Chief Executive Officer of vTv Therapeutics. “Cadisegliatin has the potential to reduce the risk of hypoglycemia, which remains a significant unmet need for people living with T1D, and we look forward to completing the study and reporting topline results in mid 2027.”

An estimated 9.5 million people worldwide live with T1D, and this number is projected to reach 14.7 million by 2040.i Despite advances in diabetes technology, significant gaps persist in achieving optimal day-to-day glycemic control,ii and hypoglycemia remains a daily burden on the lives of people living with T1D. The CATT1 study is evaluating the potential of cadisegliatin to reduce clinically significant hypoglycemia while improving overall glycemic control.

“This milestone is an important step forward for the cadisegliatin program,” said Thomas Strack, MD, Chief Medical Officer of vTv Therapeutics. “Our focus now is maintaining the quality and integrity of the study through completion and continuing the important work of advancing cadisegliatin development toward potential approval.”

About the Phase 3 CATT1 Study
Cadisegliatin as Adjunctive Therapy in Type 1 Diabetes (CATT1) (NCT06334133) is a randomized, double-blind, placebo-controlled Phase 3 study evaluating the efficacy and safety of cadisegliatin over six months in individuals with T1D 18 years or older who are on multiple daily insulin injections or continuous subcutaneous insulin infusion, along with continuous glucose monitoring. Participants were randomized to receive cadisegliatin 800 mg once daily, cadisegliatin 800 mg twice daily, or placebo. The primary efficacy endpoint will assess the incidence of level 2 and level 3 hypoglycemic events. Secondary endpoints include changes in hemoglobin A1c (HbA1c), time in target glucose range, and the incidence of diabetic ketoacidosis in cadisegliatin-treated participants compared with placebo. Additional information about the study is available at https://clinicaltrials.gov/study/NCT06334133.

About Cadisegliatin
Cadisegliatin (TTP399) is a novel, oral small-molecule, glucokinase activator being investigated in the U.S. as a potential first-in-class adjunctive treatment for T1D. In nonclinical studies, cadisegliatin acted selectively on the liver and increased the activity of glucokinase independently of insulin. These studies support clinical investigation of whether cadisegliatin can improve glycemic control through hepatic glucose uptake and glycogen storage. Cadisegliatin has been granted Breakthrough Therapy designation by the U.S. Food and Drug Administration (FDA).

Cadisegliatin is under investigation, and its safety and efficacy have not been established. There is no guarantee that this product will receive health authority approval or become commercially available for the use being investigated.

About vTv Therapeutics
vTv Therapeutics is a late-stage biopharmaceutical company focused on developing oral, small molecule drug candidates intended to help treat people living with diabetes and other chronic diseases. vTv’s clinical pipeline is led by cadisegliatin, a potential first-in-class oral glucokinase activator being investigated in a U.S. Phase 3 study for the treatment of T1D. vTv and its development partners are investigating multiple molecules across different indications for chronic diseases. ~ Acceptance into ETP enables early FDA engagement to support development of the RaniPill® platform technology ~

SAN JOSE, Calif., Oct. 05, 2026 (GLOBE NEWSWIRE) — Rani Therapeutics Holdings, Inc. (Nasdaq: RANI), a clinical-stage biotherapeutics company focused on oral delivery of biologics, today announced that the RaniPill® Capsule™ platform technology has been accepted into the U.S. Food and Drug Administration (FDA) Emerging Technology Program (ETP).

The FDA’s Emerging Technology Program, administered within the Agency’s Center for Drug Evaluation and Research (CDER), is designed to support the development and implementation of innovative approaches to pharmaceutical product design and manufacturing. Technologies accepted into the program must demonstrate the potential to improve pharmaceutical product quality and incorporate novel elements for which the FDA has limited review or inspection experience. Participation provides Rani with early, direct engagement with the FDA’s Emerging Technology Team to address technical and regulatory considerations related to the RaniPill platform, including through pre-submission meetings and potential site visits. Importantly, these interactions can address the broader applicability of the underlying technology rather than being limited to individual drug products.

“Our acceptance into the FDA’s Emerging Technology Program reflects the Agency’s recognition of the RaniPill® platform as a novel and innovative approach to the oral delivery of biologics,” said Talat Imran, Chief Executive Officer of Rani Therapeutics. “This program enables direct, collaborative access to the FDA on a technology as novel as Rani’s, and we expect that engagement to be especially valuable as we work to automate our vertically integrated manufacturing process in anticipation of broader clinical use. Combined with our rare disease, immunology, and obesity focused partnerships with Chugai, ProGen and PegBio, this milestone reflects our continued confidence in the RaniPill platform’s potential as an alternative to injectable biologics. We look forward to working closely with the FDA as we continue to advance the platform.”

About Rani Therapeutics
Rani Therapeutics is a clinical-stage biotherapeutics company focused on advancing technologies to enable the development of orally administered biologics and drugs. Rani has developed the RaniPill® Capsule™, a novel, proprietary and patented platform technology, intended to replace subcutaneous injection or intravenous infusion of biologics and drugs with oral dosing. Rani has successfully conducted several preclinical and clinical studies to evaluate safety, tolerability and bioavailability using RaniPill® Capsule technology.

Words such as “continue,” “intend,” “look forward to,” “potential,” “expect,” “will,” and similar expressions are intended to identify forward-looking statements. Continued purchases and organic accumulation drive approximately 11% growth in SOL holdings since the Company’s August 12 Earnings Update

Preliminary September 30 estimates indicate double-digit SPS growth and NAV per share growth of more than 100%, both measured from August 12

In Q3, SOL outperformed the Nasdaq-100 by 56%, and DFDV outperformed SOL by 1.5x over that same period

CHAD brings Digital Credit 2.0 to income investors: a 13% annual dividend rate supported by a productive SOL treasury

BOCA RATON, FL, Oct. 05, 2026 (GLOBE NEWSWIRE) — DeFi Development Corp. (Nasdaq: DFDV) (the “Company” or “DeFi Dev Corp.”), the first U.S. public company with a treasury strategy built to accumulate and compound Solana (“SOL”), today announced that it has added approximately 26,203 SOL to its treasury since September 28, 2026, bringing total holdings to approximately 2,564,212 SOL and SOL equivalents worth $302M.

The latest increase brings total SOL and SOL equivalents growth to approximately 11% since the Company’s Q2 earnings report on August 12. With the recently established $300 million CHAD ATM providing an additional source of growth capital, the Company believes its SOL accumulation flywheel is continuing to gain momentum.

Preliminary Q3 Estimates: Per-Share Growth Metrics and Stronger Liquidity
Alongside continued accumulation, the Company expects to report substantial growth in its per-share metrics and a stronger balance sheet as of September 30, 2026. Based on preliminary estimates, the Company expects to report the following changes as of September 30, 2026, compared with August 12, 2026:

  • Double-digit growth in SOL per share (“SPS”).
  • More than 100% growth in net asset value (“NAV”) per share.
  • Double-digit growth in total SOL and SOL equivalents.
  • A reduction in notional SOL-denominated borrowings.
  • More than 100% growth in cash and cash equivalents.

These preliminary estimates reflect roughly seven weeks of rapid progress in expanding the Company’s SOL treasury, increasing SOL exposure per common share, and strengthening liquidity.

“The DFDV ship is flying at lightning speed. We have continued buying SOL and have grown our treasury by 11% since August 12,” said Joseph Onorati, Chief Executive Officer of DeFi Development Corp. “Those gains are adding up. We expect to report double digit growth in SOL per share and more than a doubling of NAV per share since our August update. Our focus remains on making each common share represent more SOL over time.”

The Company intends to continue accumulating SOL through disciplined capital deployment and organic treasury growth, while putting its holdings to work through staking and validator operations. Its capital allocation strategy remains focused on accretive growth for common shareholders alongside prudent management of liabilities and liquidity.

The anticipated September 30 metrics are preliminary, remain subject to finalization, and may differ from the Company’s final reported results. See the forward-looking statements and supplemental metric disclosures at the end of this press release.

CHAD: First Dividend Marks a Milestone in Digital Credit 2.0
On October 1, 2026, DFDV paid CHAD’s first dividend, marking a milestone for its Nasdaq-listed variable-rate preferred stock. CHAD currently carries a 13% annual dividend rate on its $10 stated amount, equivalent to $1.30 per share annually at the current rate, with dividend payments continuing each business day when declared.

With payments underway, the Company is focused on expanding investor awareness, market access, and liquidity as it works toward establishing CHAD at its $10 stated amount and scaling it as a source of capital for DFDV’s next phase of growth.

The Company views CHAD as Digital Credit 2.0: preferred equity supported by a productive treasury. Through staking and validator operations, DFDV’s SOL holdings generate recurring rewards and fees, providing an organic source of income to support dividend obligations. The Company continues to maintain ample dividend coverage, while additional SOL accumulation can expand the treasury’s earning capacity over time.

“CHAD is fuel for the DFDV engine and central to our next phase of growth,” said Joseph Onorati, Chief Executive Officer of DeFi Development Corp. “It opens our strategy to income investors and gives us another source of capital to accumulate SOL and expand the treasury’s earning power. Our focus now is on building investor awareness, deepening liquidity, and establishing CHAD at its $10 stated amount.”

“We see a significant opportunity to scale CHAD from here,” continued Onorati. “Trading at par would position us to raise additional perpetual capital on terms that support accretive growth for common shareholders, while offering CHAD holders a compelling income stream. That is the next phase we are working toward.”

Forward Looking Statements and Metrics
This press release contains “forward-looking statements” within the meaning of the safe harbor provisions of the U.S. Private Securities Litigation Reform Act of 1995. Forward-looking statements in this press release include, but are not limited to, statements regarding expected September 30, 2026 metrics and growth in SPS, NAV per share, SOL holdings and cash balances; anticipated changes in borrowings and liquidity; future SOL accumulation and per-share accretion; capital raising and deployment, the anticipated issuance price of shares under the CHAD ATM program, the CHAD dividend rate, the declaration and payment of CHAD dividends, the intended use of proceeds, and the Company’s ability to expand its SOL treasury and increase revenue, and can be identified by words such as “anticipate,” “intend,” “plan,” “believe,” “project,” “estimate,” “expect,” “strategy,” “future,” “likely,” “may,” “should,” “will” and similar references to future periods. Forward-looking statements are neither historical facts nor assurances of future performance. Instead, they are based only on the Company’s current beliefs, expectations, and assumptions regarding the future of its business, future plans and strategies, projections, anticipated events and trends, the economy, and other future conditions. Because forward-looking statements relate to the future, they are subject to inherent uncertainties, risks, and changes in circumstances that are difficult to predict, many of which are outside of the Company’s control. The Company’s actual results and financial condition may differ materially from those indicated in the forward-looking statements. Therefore, you should not rely on any of these forward-looking statements.

Important factors that could cause our actual results and financial condition to differ materially from those indicated in the forward-looking statements include, among others, the following: (i) fluctuations in the market price of SOL and any associated losses that the Company may incur as a result of a decrease in the market price of SOL; (ii) a failure for the demand for SOL, or activity on the SOL network, to continue to develop and grow we anticipate or at all; (iii) volatility in our stock price, including due to future issuances of common stock and securities convertible into common stock; (iv) the effect of and uncertainties related to the ongoing volatility in interest rates; (v) our ability to achieve and maintain profitability in the future; (vi) the impact on our business of the regulatory environment and complexities of complying with such environment, including changes in securities laws or other laws or regulations; (vii) changes in the accounting treatment relating to the Company’s SOL holdings; (viii) our ability to respond to general economic conditions; (ix) our ability to manage our growth effectively and our expectations regarding the development and expansion of our business; (x) our ability to access sources of capital, including debt financing and other sources of capital to finance operations and growth; and (xi) the risk that the Company’s preliminary estimates as of September 30, 2026 differ materially from its final results; (xii) risks relating to CHAD, including that the dividend rate may be adjusted, that dividends are payable only when declared, that CHAD may trade below its stated amount and that staking and validator rewards may be insufficient to support dividend payments; (xiii) other risks and uncertainties more fully described in the section captioned “Risk Factors” in the Company’s most recent Annual Report on Form 10-K and other reports we file with the SEC.

As a result of these matters, changes in facts, assumptions not being realized, or other circumstances, the Company’s actual results may differ materially from the expected results discussed in the forward-looking statements contained in this press release. Forward-looking statements contained in this announcement are made as of this date, and the Company undertakes no duty to update such information except as required under applicable law.

SPS and NAV per share are supplemental analytical measures used by management to assess the Company’s treasury strategy and economic exposure attributable to common shareholders. For purposes of this release, NAV reflects the value of SOL and SOL equivalents, plus cash and cash equivalents, less SOL-denominated liabilities, outstanding principal of out-of-the-money convertible debt, other debt, and the aggregate notional amount of preferred equity. NAV per share reflects that amount divided by adjusted common shares outstanding, which includes ordinary shares outstanding, shares underlying restricted stock units and vested options, and applicable dilution from assumed exercise of in-the-money warrants and conversion of in-the-money convertible securities. Related adjustments to NAV reflect assumed warrant exercise proceeds and the removal of debt assumed converted, as applicable, to avoid double counting. SPS expresses the corresponding net SOL exposure per adjusted common share, with the SPS growth comparison applying the updated, liability-adjusted methodology and a SOL reference price to both measurement dates. These measures depend on valuation and capital structure assumptions and do not represent GAAP book value per share, liquidation proceeds, or shareholder investment returns. Preliminary estimates are not audited and remain subject to completion of the Company’s financial close and verification of underlying balances and share counts, and final results may differ materially.
