Analysis demonstrates a lower bacterial burden in breakthrough surgical-site infections, with similar proportions of multidrug-resistant organisms between treatment groups

PETACH TIKVA, Israel, Oct. 21, 2026 (GLOBE NEWSWIRE) — PolyPid Ltd. (Nasdaq: PYPD) (“PolyPid” or the “Company”), an innovative biopharmaceutical company dedicated to improving patient outcomes by elevating treatment effectiveness, right where care begins, today announced that it will present an oral presentation at IDWeek 2026 on Thursday, October 22, 2026, in Washington, D.C. The presentation will focus on an exploratory pooled analysis of microbiology from SHIELD I, SHIELD II, and D-PLEX₁₀₀ phase 2 trial involving 2,185 patients, demonstrating a lower bacterial burden in breakthrough surgical-site infections (“SSIs”) and later occurrence of breakthrough SSIs in the D-PLEX₁₀₀ treatment arm compared to standard of care (“SoC”), with similar proportions of multidrug-resistant organisms between treatment groups.

The presentation, titled “D-PLEX₁₀₀ Lowers the Microbial Burden Without Selecting for Multidrug-Resistant Bacteria: Pooled Microbiology from Three Randomized Trials (N=2,185),” will be delivered by George M. Viola, M.D., M.P.H., Department of Infectious Diseases, Infection Control, and Employee Health, Division of Internal Medicine, the University of Texas MD Anderson Cancer Center, Houston, Texas. Co-authors are Nurit Tweezer-Zaks, M.D., M.B.A., and Meidan Dvir, Ph.D.

The analysis pooled a Phase 2 trial and two Phase 3 trials to examine the bacteria present in SSIs that occurred despite preventive treatment. Wound microbiology was evaluated in 134 patients (D-PLEX₁₀₀ arm n=51, SoC arm n=83) with culture-positive SSIs. Heavy bacterial growth was less frequent with D-PLEX₁₀₀ plus standard of care than with SoC alone (16% versus 27%, p=0.034) while breakthrough SSIs occurred later in the D-PLEX₁₀₀ arm compared to the SoC (median 10.0 vs 6.5 days; p<0.001). The proportions of multidrug-resistant organisms were similar between groups, and the difference in doxycycline resistance was not statistically significant.

Across the pooled study population, no statistically significant differences were observed in non-wound infections, systemic antibiotic use, or inflammation.

“Preventing surgical-site infections remains our primary objective. Part of infection prevention is understanding the microbiology of infections that occur despite prophylaxis. This matters even more as antimicrobial resistance is a first-order global health threat1. This exploratory pooled analysis showed that compared with standard of care alone, treatment with D-PLEX₁₀₀ was associated with lower burden of bacterial growth, without evidence of increased antimicrobial resistance. Together with the later recorded onset of breakthrough SSIs, these findings suggest that D-PLEX₁₀₀ do not only demonstrate a relative risk reduction by 60% in the postoperative infection as seen in the SHIELD II trial, but also favourably impact the microbiologic characteristics and severity of infections that emerge despite treatment” said Dikla Czaczkes Akselbrad, Chief Executive Officer of PolyPid.

About D-PLEX₁₀₀
D-PLEX₁₀₀, PolyPid’s lead product candidate, is designed to provide local prolonged and controlled anti-bacterial activity directly at the surgical site to prevent surgical site infections (“SSIs”). Following the administration of D-PLEX₁₀₀ into the surgical site, PolyPid’s delivery technology, Kynatrix™, pairs with Active Pharmaceutical Ingredients, enabling a prolonged and continuous release of the broad-spectrum antibiotic doxycycline, resulting in a high local concentration of the drug for a period of 30 days for the prevention of SSIs, with additional potential to prevent SSIs caused by antibiotic-resistant bacteria at the surgical site. D-PLEX₁₀₀ recently demonstrated positive results in the Phase 3 SHIELD II trial, achieving a statistically significant 60% (p=0.0013) relative risk reduction in SSI incidence following abdominal colorectal surgery with large incisions. D-PLEX₁₀₀ received Breakthrough Therapy designation from the U.S. Food and Drug Administration for the prevention of SSIs in patients undergoing elective colorectal surgery.

Seven presentations include pediatric Phase 4 studies of AZSTARYS® (Serdexmethylphenidate/dexmethylphenidate) CII and real-world research of JORNAY PM® (Delayed-Release/Extended-Release Methylphenidate) CII in adults and impact on caregivers

STOUGHTON, Mass., Oct. 21, 2026 (GLOBE NEWSWIRE) — Collegium Pharmaceutical, Inc. (Nasdaq: COLL), a leading biopharmaceutical company focused on improving the lives of people living with serious and often misunderstood conditions, today announced that seven abstracts from its attention-deficit/hyperactivity disorder (ADHD) portfolio have been accepted for presentation at two scientific congresses this fall. The presentations span pediatric clinical studies and research examining daily functioning among adults and caregivers, reflecting Collegium’s commitment to building a broader understanding of ADHD across ages and real-life settings.

The data will be presented at the American Academy of Child and Adolescent Psychiatry (AACAP) 2026 Annual Meeting, taking place October 26–31 in Atlanta, and the Neuroscience Education Institute (NEI) Fall Congress, taking place November 5–8 in Colorado Springs.

The abstracts include Phase 4 studies of AZSTARYS® (Serdexmethylphenidate/dexmethylphenidate) CII in pediatric populations, as well as retrospective analyses of JORNAY PM® (Delayed-Release/Extended-Release Methylphenidate) CII from routine clinical practice in adults, and survey research with caregivers of children and adolescents. The real-world analyses are observational, and the investigators describe their findings as hypothesis-generating.

An estimated 15.5 million adults and 7 million children and adolescents in the United States have ADHD.1,2 Published research has associated ADHD symptom severity with greater family burden, reduced family functioning, and lower quality of life among parents.3 Subsequent work indicates that when ADHD symptoms are inadequately controlled, routine aspects of the morning can create one of the most stressful parts of the day for both parents and children.4

“People living with ADHD, and the families who support them, need care they can count on throughout the day, so they can focus on school, work, and time with the people around them,” said Thomas Smith, M.D., Executive Vice President and Chief Medical Officer, Collegium Pharmaceutical, Inc. “The clinical studies and real-world research we’re presenting this fall help deepen the understanding of those daily needs and advance how ADHD is managed.”

Before prescribing JORNAY PM or AZSTARYS, assess each patient’s risk for abuse, misuse, and addiction. Educate patients and their families about these risks, proper storage of the drug, and proper disposal of any unused drug. Throughout treatment, reassess each patient’s risk of abuse, misuse, and addiction and frequently monitor for signs and symptoms of abuse, misuse and addiction.

CONTRAINDICATIONS

Known hypersensitivity to methylphenidate, serdexmethylphenidate, or other components of JORNAY PM or AZSTARYS. Hypersensitivity reactions such as angioedema and anaphylactic reactions have been reported in patients treated with methylphenidate products.

Concomitant treatment with monoamine oxidase (MAO) inhibitors, or within 14 days following discontinuation of a MAOI, because of the risk of hypertensive crisis.

WARNINGS AND PRECAUTIONS

Risks to Patients with Serious Cardiac Disease: Avoid use in patients with known structural cardiac abnormalities, cardiomyopathy, serious cardiac arrhythmias, coronary artery disease, or other serious cardiac disease.

Increased Blood Pressure and Heart Rate.

Psychiatric Adverse Reactions: Prior to initiating JORNAY PM or AZSTARYS, screen patients for risk factors for developing a manic episode.

Priapism: If abnormally sustained or frequent and painful erections occur, patients should seek immediate medical attention.

Peripheral Vasculopathy, including Raynaud’s Phenomenon: Observe patients for digital changes during treatment.

Long-Term Suppression of Growth in Pediatric Patients: Closely monitor growth (height and weight) in pediatric patients. Pediatric patients not growing or gaining height or weight as expected may need to have their treatment interrupted.

Acute Angle Closure Glaucoma and Increased Intraocular Pressure (IOP): Patients considered at risk for acute angle closure glaucoma and IOP should be evaluated by an ophthalmologist.

Motor and Verbal Tics, and Worsening of Tourette’s Syndrome.

ADVERSE REACTIONS

Based on accumulated data from other methylphenidate products, the most common (>5% and twice the rate of placebo) adverse reactions are appetite decreased, insomnia, nausea, vomiting, dyspepsia, abdominal pain, weight decreased, anxiety, dizziness, irritability, affect lability, tachycardia, and blood pressure increased.

Additional adverse reactions (≥5% and twice the rate of placebo) in JORNAY PM-treated pediatric patients 6 to 12 years are headache, psychomotor hyperactivity, and mood swings.

In the United States, setmelanotide is indicated to reduce excess body weight and maintain weight reduction long term in adults and pediatric patients aged 4 years and older with acquired hypothalamic obesity, in adult and pediatric patients aged 2 years and older with syndromic or monogenic obesity due to Bardet-Biedl syndrome (BBS) or Pro-opiomelanocortin (POMC), proprotein convertase subtilisin/kexin type 1 (PCSK1), or leptin receptor (LEPR) deficiency confirmed by genetic testing demonstrating variants in POMC, PCSK1, or LEPR genes that are interpreted as pathogenic, likely pathogenic, or of uncertain significance (VUS).

In the European Union and the United Kingdom, setmelanotide is indicated for the treatment of obesity and the control of hunger associated with genetically confirmed BBS or loss-of-function biallelic POMC, including PCSK1, deficiency or biallelic LEPR deficiency in adults and children 2 years of age and above. In the European Union and the United Kingdom, setmelanotide should be prescribed and supervised by a physician with expertise in obesity with underlying genetic etiology.

Limitations of Use

Setmelanotide is not indicated for the treatment of patients with the following conditions as setmelanotide would not be expected to be effective:

  • Obesity due to suspected POMC, PCSK1, or LEPR deficiency with POMC, PCSK1, or LEPR variants classified as benign or likely benign
  • Other types of obesity not related to acquired HO, BBS, or POMC, PCSK1 or LEPR deficiency, including obesity associated with other genetic syndromes and general (polygenic) obesity.

— STRO-006 is Sutro’s second wholly-owned ADC to enter the clinic in less than one year —

      — Initial clinical data expected in the second half of 2027 —

SOUTH SAN FRANCISCO, Calif., Oct. 21, 2026 (GLOBE NEWSWIRE) — Sutro Biopharma, Inc. (Sutro or the Company) (NASDAQ: STRO), a clinical-stage oncology company pioneering site-specific and novel-format antibody drug conjugates (ADCs), today announced that the first patient has been dosed in its Phase 1 trial of STRO-006 in patients with a range of solid tumors. STRO-006 is the Company’s potential best-in-class integrin β6 (ITGB6)-targeting exatecan ADC with a drug-to-antibody ratio of 8 (DAR8), engineered for optimized pharmacokinetics, tolerability and a differentiated therapeutic index.

“Dosing the first patient with STRO-006 marks our second ADC to enter the clinic in less than a year and reflects the Sutro team’s exceptional execution of our pipeline strategy,” said Jane Chung, Chief Executive Officer of Sutro. “ITGB6 is difficult to target due to complex biology and is expressed across a broad range of tumors with high unmet need, including non-small cell lung and head and neck cancers. In preclinical models, STRO-006 demonstrated potent, sustained anti-tumor activity with minimal on-target toxicity, supporting our belief that Sutro’s advanced design capabilities can widen the therapeutic index and deliver a differentiated safety and efficacy profile. We look forward to advancing STRO-006 in the clinic and evaluating its potential to benefit patients.”

The Phase 1, open-label, multicenter trial is designed to evaluate the safety, pharmacokinetics and preliminary anti-tumor activity of STRO-006 in adults with advanced or metastatic solid tumors. The dose-escalation portion will enroll patients with ITGB6-expressing tumor types, including head and neck cancer, non-small cell lung cancer, esophageal/gastric cancer, colorectal cancer, endometrial cancer, urothelial cancer and breast cancer. The trial will evaluate STRO-006 across ascending dose levels beginning at 1.5 mg/kg. This starting dose is enabled by Sutro’s precise design capabilities and supported by the tolerability observed in non-human primates at doses up to 30 mg/kg.

About STRO-006

STRO-006 is Sutro’s next-generation DAR8 ADC targeting integrin β6 (ITGB6), a tumor-associated antigen expressed across several high-prevalence solid tumors. This candidate delivers a potent exatecan payload attached via a site-specific β-glucuronidase cleavable linker to a high-affinity antibody that binds to ITGB6 without disrupting TGFβ signaling. STRO-006 is designed to deliver a wider therapeutic index than conventional ADCs by enhancing selectivity, stability, and pharmacokinetics. In preclinical studies, STRO-006 demonstrated strong and durable anti-tumor activity. STRO-006 is currently being evaluated in a Phase 1 trial in patients with a range of ITGB6-expressing solid tumors.
